| Walnut,as a kind of medicine and food homologous material,it is rich in resources and high nutritional value,is called of"the four famous nuts"in the world.There is high content of glutamic acid in walnut protein,it can cause ammonia in the blood ammonia is reduced,so as to promote brain cell respiration,improve the physiological function of the brain and enhance memory.The studies have found that moderate consumption of walnuts can improve blood lipid and blood glucose levels in patients with type II diabetes,is helpful for the early stages of the insulin resistance.In recent years,our country has done some research on the walnut health care products,food and beverage and other cosmetics,but it is about walnut poly-peptide is little,in order to make better use of the resources of walnut develop the walnut products,this paper is based on the previous work,the extraction of walnut protein,poly-peptide,purification and detection of the walnut poly-peptide amino acids and the corresponding content,through in vitro experiments to evaluate its biological activity,according to the market demand,the preparation for the public demand for walnut poly-peptide products.The main results are as follows:(1)Walnut powder,walnut protein and walnut peptide,separation purification of HT-1 and HT-2:The study on the preparation of walnut meal,HT protein,poly-peptide preparation rate were 98.22%,96.71%,97.88%.The corresponding water rate were17.43%,2.02%,5.57%,ash rate were 17.58%,8.19%,8.24%,the oil in the crude powder of walnut amounted to 58.62%.Walnut powder prepared coarse texture is coarse,pale yellow,fragrant walnut flavor,HT protein is delicate,white and no strange smell,HT poly-peptide strong moisture absorption,due to the effects of protease color is brown,HT-1,HT-2 texture is very delicate,pure white,powder rate is low,in addition to containing free ammonia high volume,with arginine,tyrosine,glycine,leucine,isoleucine,methionine,glutamic acid,GABA,alanine,arginine,citrulline,tyrosine,valine and asparagine,among these amino acid relative content of HT-2 was higher than HT-1,HT-2 also contain cystine,phenylalanine,alanine,histidine,1-methyl histidine,3-methyl histidine,tryptophan,ornithine,lysine,a amino adipic acid,valine,carnosine,beta amino isobutyric acid,gamma aminobutyric acid etc.(2)The evaluation of HT protein、HT poly-peptide、HT-1 and HT-2biological activity:screening by GLUT4 transmembrane activity,glucose uptake in L6 myotubes(consumption)activity screening,MTT test anticancer inhibitory rate of HepG2 cells induced by high glucose and insulin resistance in experimental model,explore the biological activity of walnut,the alkali extraction and acid precipitation HT protein HT polypeptide,enzymatic hydrolysis,separation and purification of HT-1 after,HT-2 has hypoglycemic effect to a certain extent,the purified HT-2 is better,and has certain anti-cancer effects.(3)The preparation of walnut zinc emulsion system:according to the stability of oral emulsion and liquidity,to determine the optimal formula for soybean oil:water:emulsifier=4:2:1,the emulsifier ratio of Twain 80:Arabia:tragacanthae gum gum=8:1:1.The emulsion was prepared by dry gel method,and the amount of zinc sulfate was 5%.The walnut zinc emulsion prepared by this method has good stability and meets the quality requirements.Comprehensive study of emulsion color and flavor,taste and texture,take four factors and three levels to get optimal technological conditions of walnut zinc emulsion,determination of citric acid、VC、sucrose、sodium benzoate content were 0.2%、1.5%、5%,、0.1%.(4)Evaluation of quality of walnut and zinc emulsion Aβ1-42 N2a cell apoptosis induced by some protective effects of droplet diameter:Determination of zinc emulsion walnut is 250 m,the minimum diameter is 80 m,which drops 70%milk particle size of around 120 mum;emulsion stability at room temperature and cold storage stability are good,uniform white not layered,room temperature preservation may not exceed 2months,before drinking to shake,chilled taste better.Using flame atomic absorption spectrometry,the content of zinc in three kinds of walnut zinc emulsion was 4.22,4.64and 4.72 g/ml respectively.The results of in vitro cell experiments showed that walnut zinc emulsion can protect N2a cells from apoptosis induced by Aβ1-42 and walnut peptide zinc emulsion has obvious improvement effect. |
