Applications Of Fluorene Based Water Soluble Conjugated Polymers In Diagnosis, Imaging And Therapy | | Posted on:2018-06-21 | Degree:Master | Type:Thesis | | Country:China | Candidate:G S Zeng | Full Text:PDF | | GTID:2321330536979691 | Subject:Bioelectronics | | Abstract/Summary: | PDF Full Text Request | | In recent years,water soluble conjugated polymers(WSCPs)have been widely used in the biological field due to their unique optoelectronic effects.According to the application,the application of water-soluble conjugated polymers in biology is divided into three parts: diagnosis,imaging and treatment.However,there are still some disadvantages such as low quantum efficiency,poor biocompatibility and poor response.In order to solve these problems,we have developed a series of novel water soluble conjugated polymer probes.Because of its excellent optical properties and easy modification,poly(fluorene)is an ideal material for water soluble conjugated polymer probes.In this paper,the preparation of this paper introduces three kinds of different types of fluorene based water soluble conjugated polymer materials,and explore their biological properties and its application in the detection of thiols from hypoxic imaging and photothermal therapy in three aspects.(1)Developed a side chain disulfide bond containing water-soluble fluorene based conjugated polymers thiol detection probe,through the replacement of effects of sulfhydryl disulfide,changing the probe water solubility,detection of thiol detection "Turn-off" effect.At the same time,the probe can realize quantitative and highly selective detection of thiols and show good detection effect in cells.(2)Introduced a kind of azobenzene(AZO)side chain water soluble polyfluorene conjugated polymer hypoxia imaging probe based on fluorescence resonance energy transfer(FRET).Most cancer cells are in varying degrees of hypoxia,because the absence of oxygen,the cells will produce azo reductase,AZO can be interrupt,FRET effect disappeared.Good FRET effect of AZO and Polyfluorene made the imaging a "Turn-on" effect,and the probe exhibit good imaging efficiency in hypoxic cells.(3)The synthesis and thermal properties of 1,4-diketo-pyrrolo-pyrrole(DPP)doped water soluble near infrared Polyfluorene was investigated.After DPP doping,the absorption and emission of the materials are in the near infrared region,which makes the material excellent candidate for in vivo imaging.The photo-thermal effect of the material in aqueous solution was investigated under laser irradiation,and the probe shows greatpotential in photo-thermal treatment. | | Keywords/Search Tags: | Water Soluble Conjugated Polymers, Polyfluorene, Disulfide, Thiol Detection, Azobenzene, Hypoxia Imaging, 1, 4-Diketo-Pyrrolo-Pyrrole, Photo-Thermal Effect | PDF Full Text Request | Related items |
