In this study,we used Auricularia cornea as test spawn to study the production process and inoculation process of new spawn.At the same time,the preservation experiment of new spawn was studied,and the extracellular enzyme activity in spawn and preserved spawn was studied.Which provided the scientific basis for the study of the fungus species.The conclusion is as follows:(1)The optimum formula is 94.5% of the hemp rope,2% of the wheat bran,1% of the glucose,the optimum amount is 10 cm / root,the optimum water content is 50%,the pH is 6,the hemp diameter is 6mm,Potato juice 1%,lime 1%,gypsum 0.5%.(2)Vaccination process of new type of spawn: In the sterilization work inoculation of the inoculation work carried out the most ideal,the use of new spawns than the branches of spawn spawns of spawn bag pollution rate of 0.94%-2.74%;new spawns of spawn The growth rate of mycelial growth rate was higher than that of other solid species.The average growth time was only that of liquid Spawn slow 2d for 26 d,with the liquid spawn yield of almost 748.6g / bag,more than the wood species and branches of the spawn 1.93 g / bag and 1.17 g / bag.The germination rate and mycelial growth were better at 10 ℃ than that of other treatments.The preservation effect of woody and new strain was better than other treatments.(3)In the extracellular enzyme of mycelium,ligninase(laccase)was found to decrease enzyme activity with the increase of time during the growth stage of mycelium.Cellulase and hemicellulase were incubated with the time of mycelial growth Increased activity of enzyme increased gradually,amylase activity is higher,in the mycelial growth stage activity gradually increased.On the whole,the enzyme activity of the new spawn and the wheat variety was higher than that of the other two spawns.The activity of the enzyme was similar to that of the untreated spawn,but the activity of the enzyme decreased with the increase of time and temperature.The enzyme activity of the new spawn and the wheat seed was better than that of other treatments at 10 ℃.(4)In the analysis of the correlation between the extracellular enzyme activity and the growth period traits of the fungus and the preserved fungus,the laccase activity in the spawn was positively correlated with the yield.The activity of carboxymethylcellulose and the activity of hemicellulose The germination time,growth potential,growth rate and yield of mycelium were correlated,and the activity of amylase was related to the time of primordia and maturation time of fruiting body.The activity of laccase in the preserved spawn was positively correlated with the germination time and the number of bags per day.The activity of carboxymethylcellulose and the germination time and growth potential of the mycelium were related,the hemicellulose and the survival rate and germination time There was a correlation between amylase activity and full bag days.
