In this paper,VitisflexuosaThunb fermentation broth in the fermentation process as the research object,were investigated by capillary column Gas Chromatography determination of ethanol in fermentation liquidVitisflexuosaThunb,VitisflexuosaThunb to determine the content of polysaccharide in fermentation liquid to improve and optimize the phenol sulfuric acid method to establish a method for determination of polysaccharide content in VitisflexuosaThunb fermentation system the.With VitisflexuosaThunb fermentation liquid alcohol content and polysaccharide content changes,further investigated the application of near infrared spectroscopy in VitisflexuosaThunb fermentation process,discussed the influence of VitisflexuosaThunb fermentation liquid modeling of near infrared spectra and the selection of the establishment of the VitisflexuosaThunb fermentation liquid spectrum processing method and instrument parameters,spectral range the model was validated with the research.In order to provide some theoretical data for on-line monitoring and testing of fermentation process.By using capillary Gas chromatography to determine the content of VitisflexuosaThunb ethanol in fermentation liquid,because ethanol is a chemical substance highly polar and volatile,we were chosen in chromatographic column and the internal standard,the final detection method of HP-INOWAX capillary column and n-propanol as internal standard.To solve the separation degree and peak effect and column efficiency low,so as to improve the accuracy and precision of the analysis results.Optimization of phenol-sulfuric acid method(Phenol Sulfuric Acid Method)to determine the content of VitisflexuosaThunb determination of polysaccharide in fermentation liquid.The experiment based on VitisflexuosaThunb fermentation broth was precipitated with lead acetate pretreatment,phenol sulfuric acid method pigment to solve the interference problem.The optimum conditions were determined by the amount of phenol,water bath temperature and bath time.This method is based on the previous research results,a method used for the determination of VitisflexuosaThunb polysaccharide in fermentation liquid content,provide theoretical data for the determination of polysaccharide VitisflexuosaThunb and follow-up study.The effects of different spectral pretreatment methods,instrument parameters and spectral range selection on the near infrared model of fermentation liquid were studied by means of transmission spectrum acquisition.The results show that the optimal model is established respectively for the number of resolution 8cm-1 and scanning for 64 times,through the rational selection and selection of light pretreatment spectral region,the establishment of a quantitative model of alcohol and polysaccharide in fermentation liquid VitisflexuosaThunb,is suitable for predicting unknown samples of fermentation liquid.Infrared spectra of the original graph reflects the spectrum of main chemical components in the fermentation broth of VitisflexuosaThunb absorption peak and there is a linear relationship between the chemical group.Chemometrics spectral processing and spectral interval selection and analysis of the original spectra by using near infrared spectroscopy,by comparing a derivative,two order derivative with curl and smooth spectrum processing method,using F,D+N,D treatment conditions,can effectively eliminate the scattering of light and stray light and the instrument response baseline near infrared spectroscopy and spectral drift effects caused by the repeated problems,establish alcohol qualitative model using discriminant analysis method for fermentation period and stable period of fermentation broth VitisflexuosaThunb discriminant analysis,discusses the relationship between alcohol fermentation and polysaccharide.A neural network model of the Vitisflexuosa Thunb alcohol fermentation,the influence of polysaccharide,alcohol concentration,fermentation temperature,p H value,humidity and other parameters on the fermentation of alcohol fermentation is discussed,the results show that the polysaccharides content had the greatest influence on ethanol fermentation,the weight is greater than 45%,the size effect of polysaccharide,p H,fermentation temperature,fermentation the humidity of the environment.The above research and discussion of the content is suitable for the prediction of unknown samples,rapid determination of alcohol fermentation liquid in the fermentation liquid and the content of polysaccharide and determine the fermentation period for online monitoring,impact assessment of fermentation parameters provide theoretical data support.
