Objective: In view of the protein contained in Periplaneta americana.The extraction process of Periplaneta americana protein was carried out by various methods of extraction,screening and optimization.To provide technical support for the efficient extraction of protein from Periplaneta americana.Bovine serum albu min was used as a reference.The protein content of Periplaneta americana in the sample was deter mined by Coomassie brilliant blue method.The protein content of Periplaneta americana in the sample was deter mined by Coomassie brilliant blue method.The amount of protein extracted under different conditions was compared.The use of drug replacement methods.The effects of Periplaneta americana protein on woundhealing in injured mice were observed and recorded.It can provide reference for further research of American cockroach in food and medicine.Method:The protein of Periplaneta americana was extracted by alkaline extraction,salt extraction and trypsinhydrolysis.The effects of different extraction methods on extraction rate were compared.The optimum extraction conditions of Periplaneta americana protein were deter mined.The protein content of the extracted of Periplaneta americana using Coomassie brilliant blue method were identified.The use of Periplaneta americana protein as a drug was used to test drug replacement in mice.Result:This experiment used three methods to extract protein from Periplaneta americana.Among of these,when the alkali concentration was 1.5 %,the extraction temperature was 80 ? and the extraction time was 1h of alkali extraction,the protein extraction rate of Periplaneta americana was thehighest.When the salt concentration was 1 %,the extraction temperature was 40 ?,and the extraction time was 2h of salt extraction,the protein extraction rate of Periplaneta americana was thehighest.When the protein extraction rate of Periplaneta americana washighest when the sample ratio was 0.6 %,the enzymolysis temperature was 45 ?,and the enzymolysis time was 2h of trypsin extract.Comprehensive experimental proof.The extraction rate of Periplaneta americana protein by trypsinhydrolysis was 58.87 %,higher than other methods.The optimum extraction conditions are A3B2C1,that is,the ratio of enzyme to sample is 0.6 %,the extraction temperature is 45 DE G C,and the extraction time is 1h.The extraction efficiency of Periplaneta Americana is thehighest,and the method is the best.The woundhealing test in mice showed that,the protein extracted from Periplaneta americana can promote woundhealing in mice.Moreover,the time of woundhealing in mice using Periplaneta americana protein was nearlyhalf shorter than in normal conditions.Conclusion : The extraction experiments of Periplaneta americana protein showed that the extraction rate of different extracts varied in different ways.Trypsinhydrolysis is the best method for protein extraction of Periplaneta americana.And the experimental conditions can be controlled.The obtained Periplaneta americana protein can effectively promote thehealing of wound in mice,andhas the prospect of the development of medicinal products.
