Considering the vital biomolecule and ions in living organisms,the accurate,rapid and portable bio-analytical methods should be developed which has important significance for the treatment and prevention of the disease.A positivelycharged perylene derivative with excellent optical properties was employed as a probe for the detection of alkaline phosphatase based on its aggregation induced by polyphosphoric acid.Furthermore,Cu2+was detected based on the fluorescence quenching of the nano-particles of perylene derivative.Based on polyphosphoric acid inducing positively charged perylene derivative probe aggregation,a fluorescence turn-on method for detection of alkaline phosphatase was designed and developed.Foremostfluorescence was quenched by aggregation of probe-1 as induced by polyphosphate.However,the addition of alkaline phosphatase hydrolyzed the polyphosphoric acid and restored the fluorescence by releasing probe.The detection limit was observed to be 0.5 mU/mL.In the second stage nanoparticles of negatively charged perylene derivative was synthesized.Those were found chemically stable,exhibiting good water dispersity and strong luminescence of excimers.Based on the quenching of nanoparticles fluorescence by Cu2+,this thesis developed a method for Cu2+detection with 20 nM limit of detection.
