Cervical cancer is one of the most common cancers,which threats to women’s physical and mental health seriously.Previous studies showed that baicalein(BAI)had obvious effects on U14 cervical tumor-bearing mice models and He La cells.However,the use of BAI is limited due to its low bioavailability.Hence,it is very important to deliver the BAI to the tumor site with a targeting delivery system.In this study,BAI-loaded nanoliposomes(BLs)were first prepared by thin film hydration method.Furthermore,Chitosan(CS)and folic acid(FA)were selected as the modifying materials to coat BLs.During the coating process,the folic acid–chitosan(FA-CS)conjugates were synthesized and characterized using infrared spectrum analysis.The appearance of the peak at 1607 cm-1 in the spectrum of FA-CS was a typical vibration of FA.New active targeting drug delivery systems for the delivery of BAI,CS-coated BLs(CBLs)and FA-CS-coated BLs(FCBLs),were constructed by CS and FA-CS coating on the surface of BLs by ion adsorption and electrostatic interaction.The transmission electron microscopy(TEM)results showed that BLs,CBLs and FCBLs were spherical.Malvern laser particle size analyzer was used to measure the particle size and Zeta potential of the liposomes.The size distributions of the three kinds of liposomes were uniform and the average particle size was all between 100 nm-200 nm.The encapsulation efficiencies(EE)of BLs,CBLs and FCBLs were all higher than 80%.The BLs was negatively charged,and the charge was reversed after CS modification.Stability tests of drug storage showed that there were no significant changes in average particle size,polydispersity index(PDI),Zeta potential and EE after 4 weeks of storing at 4 ℃ for the nanoparticles.There were slow release effects for BAI in all of the three kinds of liposomes.In addition,the anticancer effects of the three kinds of liposomes were studied by SRB assay and U14 cervical cancer bearing mice model in vivo and in vitro respectively.The results showed that all of the three liposomes enhanced the antitumor effect of BAI,and FCBLs exhibit the highest tumor inhibition effect by the binding of FA and FA receptor,and the adsorption of CS.In conclusion,the study suggested that FCBLs would serve as a potential drug delivery system in future cancer therapy.
