As an important component of the human body,collagen is a type of biological macromolecule structure(or functional protein)synthesized by cells.With many excellent biological properties owing to its unique triple-helical structure and distinctive function as nutritional protein,collagen has aroused increasingly widespread concerns of scholars.At present,the collagen sold in markets is mainly derived from mammals.But in recent years,its security has been questioned due to the outbreak of mammalian diseases and the growing prevalence of vital zoonoses,leading to the worldwide search for safer collagen source.In this case,collagen derived from aquatic products has drawn wide attention because of its advantages of abundant raw materials,low antigenicity,low sensitization,high solubility,no risk of infectious diseases and no religious barriers.In this paper,we mainly discussed the study on the preparation and property analysis of fish source collagen and bio-materials constructed through its self-assembly,including the following three parts:In the first part,the paper reviewed the source,composition,structure,extraction methods,physicochemical properties,biological activity,application,research progress and other aspects of fish collagen,and then focused on the application of collagen's self-assembly property in medical biomaterials and proposed the main research contents and research directions of this subject according to the current research status.In the second part,a new preparation technique of fish source type-? collagen was established through integration and innovation,and the performance of this technique was analyzed.In this study,the skin of Tilapia,the by-product obtained in the process of aquatic production in China,was chosen as the research object for development.And acid soluble collagen(ASC)and protein soluble collagen(PSC)were prepared by applying biochemical methods like acid extraction method,and enzyme extraction method.Then,the extraction rate of collagen was employed as an index to explore the extraction technique of type-? collagen.Based on this,the study of structure identification,molecular weight determination and physicochemical proper ty analysis for medical use were conducted.In the third part,research and development of new biomaterials were carried outusing the prepared bioactive type-? collagen as the raw material.Next,collagen fibrils was constructed through the self-assembly of type-? collagen.And the fiber recombination technology was selected to develop absorbent cotton,a hemostatic absorbent material for cavity packing,and finally explore a high value-added industrial chain with low-priced protein source.In the property determination tests,the prepared absorbent cotton exhibited complete triple-helix structure with porous net structure constructed by linear fiber,providing excellent supporting environment for cell adhesion and cell proliferation,which contributed to its outstanding hemostatic performance and served as the basis and reference for further research and development of absorbent cotton as hemostatic materials for nasal and auricular packing as well as hemostasis.
