Nano silver is a conjunction of silver atom about 1-100nm. Silver nano particles use wide in our common life with antimicrobiology, such as the area of medicen, food package.Under low dose, silver nano particles are antibacterial agents but if under high does, several cell lines were considered to be apoptosis through peroxide the membrane lipid, disruption the mitochondrial membrane potentials and oxide the mitochondrial electron transport chain complex, if nano silver treated cell, ros in the cell were elevated and induce cell death.Mitochondrial DNA polymerase gamma is a unique DNA polymerase in mitochondrial and it is a key enzyme to keep dna fidelity and mitochondrial balance. Mitochondrial damage is a key factor in induce cell apoptosis and disruption mitochondrial dna integrity are also a key results of mitochondrial disfuction. In this paper, we were the first put forward which nano silver would conjugated with mitochondrial dna polymerase gamma and then damage the cell function and how this binding would work.In order to speculate the namo silver toxicl,we measure the phnotype after the cell treated with nano silver;then we use MTT assay to measure the cell survival;next,Real time PCR measure the express level in mitochondiral DNA polymerase gamma;the most important experiment is the nano silver pull down with mitochondrial polymerase gamma.In this paper, the first work is measuring Hela cell damage in various nano silver concentration, and observe the cell morphology and cell survival. In our experiments we conclude that under high dose silver nano particles the cell would change their shape from skin to radiate physiology, but if cell under low dose concentration, cell would have no transformation under view. MTT experiments show that under high dose concentration cell would be death. This results would converse to previous consideration that silver nano particles were antibacterial agent and would be used in human common life. Aimed at concerning mitochondrial DNA polymerase gamma gene transcriptions, we prepared real time PCR experients and the results show that the cell under low dose silver nano particle concentrations the aim genes were up express and conversely, if the concentrations were high, genes of mitochondrial dna polymerase gamma would depress expression. Next, we asked would nano silver bind with mitochondrial dna polymerase genes directly and influence cell function. In vitro experiments of nano silver and mitochondrial protein lysis and experiments of nano silver with pure protein of mitochondrial DNA polymerase gamma were performed and the results suggest that nano silver particles would binding directly with our proteins.
