| Ginseng is a traditional Chinese medicinal herb.The major bioactive constituents of ginseng is ginseng saponins.Have high medicinal value.Minor saponins C-K and Rhl is the body metabolism of panaxadiol and Panaxatriol saponins respectively,It does not exist in nature,or in extremely small.But there is a marked pharmacological effect.The method of transformation ginsenoside has a lot,but they have a lot of problems,for example poor specificity,lot byproduct,environmental pollution and so on.Microorganism method shows great advantage,such as simple operation,low price.So we separate microbial strains from the soil of the Forest Senate.Identify effective strains of transformed ginsenosides.Study on physiological characteristics and aspects of culture conditions of Microbial strains.In this study,we isolated 139 kinds of microorganisms from the soil and screened 75 β-glucosidase producing strains.After that,GS33 和 YD18 were found to convert ginseng saponins to C-K and Rhl efficiently.We had ITS sequencing of high performance strains,and had 16S rDNA sequencing.According to the sequencing results to construction phylogenetic trees,and combination with morphological to analyze strain GS33 as Penicillium simplicissimum,strain YD18 as Penicillium.Through optimization of conditions,we found that the strains transformat ginseng root saponins most completely and produce the most amount of C-K in the medium of water,pH 3.0.Quantitative analysis by HPLC,we found that the strain GS33 generate the yield of C-K and Rhl were 14%and 25%,separtately.Optmization of conditions of YD18 is in the medium of water NB LL and R2A,pH 4.0-7.0.Quantitative analysis by HPLC,we found that the strain GS33 generate the yield of C-K and Rhl were 11%and 18%,separtately.When the strains transformat ginseng-lixivium,optimization culture time of ten strains,found that training 11 d-13 d is the best,meanwhile MTT experiment was on transformation of product,The results show that the product of the transformation of strain GS33 significantly reduced the activity of cancer cells,whent election HepG-2,different dose did not show a very different.When election ES-2,transformation products of strain GS33 inhibition rate was 58.42%,When the dosage was 800 ug. |
