| With the rapid development of culture and material,consumer have higher requirement of food color,flavor,nutrition and safety.Sulfide flavor is the maximum kind of sulfur flavors,it has an important role in improving the flavor of food.But currently study of anti-tumor activity and antimicrobial activity on sulfide flavors is still relatively rare.Cytotoxicity induced by 19 kinds of sulfide flavors was investigated in human leukemia HL-60 and Jurkat cells.The structure-activity relationship was further analyzed according to the comparative studies,and the mechanism involved in cytotoxicity was primarily studied.The results showed that difurfuryl disulfide(DFDS),bis(2-methyl-3-furyl)disulfide(BMFDS)and methyl 2-methyl-3-furyl disulfide(MMFDS)had stronger cytotoxicity in HL-60 and Jurkat cells than the other flavors.These three sulfide flavors could induce DNA fragmentation and cell viability decrease of HL-60 and Jurkat cells,and it had dose-effect relationship.According to the structure-activity relationship of sulfide flavors,it is concluded that the number of sulfur atoms and furyl play important roles in cytotoxicity of sulfide flavors in HL-60 and Jurkat cells.In order to further elucidate the mechanism involved in cytotoxicity of these three sulfide flavors,ROS production in Jurkat cells was determined.These results suggest MMFDS and BMFDS possibly induce cytotoxicity by ROS production in Jurkat cells.In order to elucidate the mechanism involved in cytotoxicity of these two sulfide flavors,Jurkat cells were pretreated with antioxidant GSH and NAC,respectively,and the effects of sulfide flavors on cell DNA fragmentation was further investigated.The results showed that the antioxidant had significant protective effect of on cytotoxicity induced by BMFDS and MMFDS in Jurkat.These suggest that MMFDS and BMFDS really induce cytotoxicity by ROS production in Jurkat cells.The antibacterial activity of thirteen sulfide flavors to the four tested bacteria(Staphylococcus aureus,Listeria monocytogenes,Escherichia coli,Vibrio Parahaemolyticus)was determined by measuring the diameter of inhibition zone.These thirteen sulfide flavors all have antibacterial activity,3-(methylthio)propionate has the best effects of antibacterial activity.Aldehyde may greatly improve antibacterial activity of sulfide flavors.Further determination of 3-(methylthio)propanal minimum inhibitory concentration(MIC)in four tested bacteria,results showed that MIC for all tested strains≤625mM,which Vibrio Parahaemolyticus lowest MIC was 2.44mM. |
