Synthesis of α-tocopherol succinate by enzymatic immobilized lipase was studied in this paper,the following outcomes are demonstrated.The influence factors of esterification process of α-tocopherol succinate by enzymatic immobilized lipase(in the shacking table)were investigated.Lip400 of lipase was best enzyme type by screening experiments among several types of immobilized lipase.The reaction conditions were as below: α-tocopherol concentration was 10g/L,the mole ratio of succinic acid/α-tocopherol was 5:1,lipase(mass quantity)was the 15% of α-tocopherol,reaction remperature was 30℃,shacking table rate was 200 rpm,reaction time was 6 hours,α-tocopherol succinate yield rate was 80%.In the fixed-bed(continuous reactor),the influence factors of esterification process of α-tocopherol succinate by Lip400 of lipase were investigated.The reaction conditions were as below : α-tocopherol concentration was 8 g/L,the mole ratio of succinic acid / α-tocopherol was 2:1,feed rate was 0.5 mL/ML/min,reaction remperature was 30℃,α-tocopherol succinate yield rate up to 86%.To improve the yield rate,the reaction kinetic mechanism of esterification process of by enzymatic immobilized lipase were investigated.The following outcomes are demonstrated as below: α-tocopherol succinate enzymatic esterification kinetic mechanism was matched with Michaelis-Menton equation.Ping-pong Bi-Bi kinetic mechanism well expound this enzymatic esterification.The corresponding kinetic equation and parameters are reached.To furtherly improve the yield rate,the influence factors of this esterification process under the reactive distillation were investigated.The following outcomes were demonstrated as below: under the same reaction conditions,the yield rate will increase by respectively enhancing the succinic acid volume,lipase volume and solvent volume.The reaction conditions are: the mole ratio of succinic acid/α-tocopherol was 1.05:1,lipase mass quantity was the 10% of α-tocopherol,reaction remperature was boiling point of the reaction system,α-tocopherol succinate yield up to 98%. |
