In the field of tissue engineering, wound infection caused by microbial and bacterial contact with human tissues has become serious problem.Bacterial infections can cause harder wound healing, and result in a series of inflammatory reactions, which will increase the patients' pain.In addition, the bacterial adhesion of implant surface and the growth of biofilm have been regarded as a problem in the biomedical field. The formed biofilm will protect bacteria from antibacterial agents, leading to the bacterial drug resistance, which will bring about tissue damage and issues of dysfunction. Therefore,it is necessary to study various methods to inhibit bacterial adhesion and bacterial growth so as to prepare biocompatible and non-toxic coating materials without immune rejection. In this work, dopamine and its derivatives can be used as crosslinking agent and adhesive materials for the study of biomimetic antibacterial materials. It is of great significance in the field of biomedicine and materials science to further study the preparation of the antibacterial hydrogels and antibacterial polymer membranes based on dopamine. The specific work is as follows:1.Bilayer bioadhesive hydrogels with antibacterial and antifouling activityWe successfully synthesized bilayer bioadhesive hydrogels P(PEGDA-co-SBMA)+P(PEGDA-co-DMA-co-SBMA)with antibacterial and antifouling activity by photopolymerization method. The successful fabrication of the samples was confirmed by Fourier transform infrared spectroscopy(FTIR).Then, the swelling properties and rheological properties of the gels were investigated.Antimicrobial and anti-adhesion properties of S.Aureus and E.coli were investigated by antibacterial experiment in vitro. And we selected bovine serum albumin as a template in the protein adsorption experiment, and we studied the anti-protein adhesion ability of gels. Finally, the results of the antiplatelet adhesion properties of the polymers and the toxicity to HL7702 cells revealed that the materials have good biocompatibility.2.Bio-inspired adhesive polymer coatings containing dual antimicrobial activityWe successfully synthesized bio-inspired adhesive polymer coatings containing dual antimicrobial activity by photopolymerization method. The synthesis of the target product was confirmed by Fourier transform infrared spectroscopy(FTIR).Then, the swelling and rheological properties of the gel were investigated. In order to enhance the antibacterial activity,P(DMA-co-CA-co-MEDAB)-Van polymer film was synthesized on the surface of titanium alloy(TC4,Ti)by adding antibiotics. Finally, the antibacterial,anti-adhesion,blood compatibility, cell compatibility properties of hydrogels and polymer coatings were studied.
