In this paper,a two-step culture method was applied to the production of lutein by Sc.633,and the effects of nutrient conditions and environmental factors on the biomass and lutein production were studied.The results showed that mixotrophic cultivation could promote the growth of biomass and the accumulation of lutein,and the glucose was the most suitable organic carbon source.When the glucose concentration was 15 g·L-1,the synthesis of lutein was the most favorable.Inoculation amount of 20%ensure that the algae can adapt to the concentration of glucose in order to achieve higher rates of growth.When the initial pH was 8,the biomass and lutein yield reached the maximum.The stress condition could further promote the accumulation of lutein,and the addition of nitrogen deficiency,phosphorus deficiency,high temperature and oxidant had different effects on the growth of biomass and the accumulation of lutein.The two-step cultivation method of"mixotrophic-autotrophic" mode was established by optimizing the various conditions to increase the biomass growth under autotrophic conditions and the use of stress conditions to accumulate a large amount of lutein.After repeated experiments,the highest lutein yield was 31.8 mg·L-1,and the content of lutein in unit mass was 12.92 mg·g-1.Compared with the traditional one-step autotrophic method,the content was increased by 38.9%.Two-step cultivation has greatly improved the yield of lutein,which is of great application value.The optimum conditions were determined by single factor experiments and orthogonal experiments.Methanol:dichloromethane = 3:1 was the best extractant,the optimum ultrasonic power was 135 W,the optimum ultrasonic time was 20 min,the best ratio of material to solvent was 2:1.Ultrasonic extraction method can save more solvent than the traditional solvent extraction method,with a short time,extraction efficiency,energy saving and environmental protection,it is worth developing.
