Abstract/Summary:
Low phenylalanine protein is a raw material of preparation the special diets for the phenylketonuria,so preparation of low phenylalanine diets caused widely attention.But there are still some problems,such as the content of phenylalanine is not up to the standard,most of the food are synthetic and lack of security.Because rice protein has low allergy,high biological titer,and the phenylalanine content is relatively low,this study selects the rice protein as raw material.With the research on solubility of rice protein laid a good foundation for the efficient removal of phenylalanine,determine the method to improve the solubility of rice and different physical treatment to explore the influence of its protein properties.Determines the full free phenylalanine and fast free phenylalanine method,has been clear about the dynamic adsorption to removal the phenylalanine,and analyzed the properties of rice protein changes in the process of phenylalanine removal,to the preparation of low phenylalanine rice protein makings.On the basis of the scale-up experiments,determine the development of low phenylalanine of meat food formula.And provide the reference for the preparation of low phenylalanine special food research and product development.Through the analysis of the rice protein components,has been clear about the reasons of the poor solubility.According to Osborne grading method,rice protein gluten content is the highest(92.43%),albumin,globulin,gliadin content is low as 3.66%,2.19% and 1.72% respectively.As a result of higher gluten content,the phenylalanine content is 90.20% of the total content of phenylalaninef in gluten.Gluten with high hydrophobic,the solubility of rice protein is bad.It was cleared that the ultrasonic cell crushing is the best pretreatment way: through a study of the rice protein structure,you can see,scanning electron microscopy(SEM)show that ultrasonic cell crushing processing surface structure of protein is more obvious than the other methods,makes the protein structure is loose and connection between protein deep bond rupture degree;Fourier infrared testing secondary structure observation of secondary structure changes significantly,the hydrophobic forces also suggests that it has superiority to the solubility.The improvement of the solubility of rice protein for the efficient removal of phenylalanine laid a good foundation.Through the full digestion and fast digestion process research,determine the two methods of free phenylalanine.Rapid enzyme solution: using neutrase with pronase compound enzyme solution,adding neutrase 5600U/g to 1% rice protein solution at 50℃,react time was 2h.For pronase,the enzymatic time was 4h,the temperature was 35℃.The best amount of pectinase was about 98U/g protein.Through enzymatic hydrolysis,the Free-Phe content can reach 80%.Enzymolysis time reduced to 4 h,enzyme solution with high efficiency.Full enzyme solution: using chymotrypsin with pronase compound enzyme solution,adding chymotrypsin 7200U/g to 1% rice protein solution at 50℃,react time was 6h.For pronase,the enzymatic time was 4h,the temperature was 35℃.The best amount of pectinase was about 98U/g protein.Through enzymatic hydrolysis,the Free-Phe content can reach 92.25%.Enzymatic hydrolysis time reduced from 18-24 h to 11 h,but the digestion time is too long.Compared the static adsorption and dynamic adsorption of two kinds of phenylalanine content in the form of rice protein,and determined the best way to adsorption.With adsorption,the final Phe content was 0.13%.The surface hydrophobicity,relative molecular weight,amino acid sequence of rice protein hydrolysate was significant changes.The structural characterization of rice protein standed for the Phe had been set free.The acute toxicity experiment,the low phenylalanine rice protein hydrolysate non-toxic,formula can be used for low phenylalanine protein diets makings.To study the scale-up preparation of low phenylalanine rice protein.Adopt the method of fast free phenylalanine scale-up experiments,production magnification of 100 times.Using adsorption process of simulated moving bed chromatography removal phenylalanine,capacity of 4.32 L/d,enlarge 26 times.Large industrial simulated moving bed chromatography removal phenylalanine,processing of 40000 L/d,enlarge 260000 times.Preparation by dry and wet preparation of low phenylalanine,meat food technology research,the best process and formula is determined.Low phenylalanine diets is the preparation of low phenylalanine makings,glucose syrup,triglycerides,soy lecithin,low poly fructose,edible salt,fish oil,vitamin A,vitamin B1,vitamin B2,vitamin C,vitamin D,vitamin E in different ratio.Determine the process of preparation low phenylalanine special food with method of the formula.Doing the sensory evaluation and cost analysis,of prepared product with the dry method and wet method.And the wet method is superior to the dry preparation is determined.In this research,studied the combination of physical and biological enzymatic treatment to improve solubility of rice protein,by using the combined method of digestion and absorption of rice protein removal of phenylalanine in the preparation of low phenylalanine rice protein makings.At the same time,respectively to scale-up of enzyme desorption,and determine the preparation of low phenylalanine,rice protein diets of formula,for low phenylalanine,provides the theory basis for the preparation of meat food,to benzene acetone urine sickness patients brought the Gospel.
