A epimedium polysaccharide conent analysis method-3,5-dintro salicylic acid colorimetic method(DNS method)is established,application of this method for the determination of epimedium polysaccharide content,this experiment is the first attempt.Compared with other spectrophotometric,the determination of epimedium polysaccharide not only do not use sulfuric acid,used in the stability of the reagents,and can be measured directly after removal of the monosaccharide composition interference,the result are accurate.The experiment while trying to create a DNS method for the determination of epimedium polysaccharide DNS method investigation of the reaction conditions and the results can be showed that the method is simple,the results are accurate,reproducible and can be used for the determination of prositutionsheep hopane polysaccharide.In this study,a complete epimedium polysaccharide preparation routes,the first using the boiling method to extract the polysaccharide,and draw on the preparatory work of the TASK Force,membrane separation before the experiment a step-by-step concentration,activated carbon adsorption,muli-step processing,effectively removed most of the impurities,reducing the degree of membrane fouling,membrane flux;one of the experimental microfiltration as pretreatment of ultrafiltration,and througu single factor experiments,orthogonal to determine the optimum ultrafiltrationpross conditions,the final application of this condition,the preparation of epimedium polysaccharide by enzyme addition to the protein,to obtain Epimedium polysaccharide of high purity.Use of membrane separation technology to deal with Chinese herbal extract,the phenomenon of membrane fouling in the microfiltration process is still more common,further research on this experiment on membrane fouling in the microfiltration pretreatment of the feed solution permeate flux decayand exploring effective solutions.Emphasis on the extent of the high concentration of solid to liquid at constant pressure conditions,the transformer conditions and different transformer time membrane fouling of the experiment,epimedium polysaccharide and preliminary experiments to conduct a preliminary investigation,and last through the pine nut shell polysaccharide experiment concluded,transformer operation of the high concentration of solid to liquid and transformer time fixed for 0.5h,can effectively control the permeate flux decay,reduce membrane fouling,pre-treatment in the multi-step membrane separation is a very important reason.UF experiments,respectively,to examine the size of the different pressure,concentration,temperature of membrane flux,the results showed that:the permeate flux with pressure,temperature increase,decrease with increasing feed concentration,and the theoryknoledge consistent,effective verification of theoretical knowledge,and the orthogonal experiment,the optimum conditions for the preparation of epimedium polysaccharide.UF experiments were prepared polysaccharide color,high quality,and green,showing its broad into production prospect.In this study,epimedium polysaccharide Enzymatic Removal of Protein research,design-Response Surface Optimization of epimedium polysaccharide Enzymatic Removal of Protein.Amount of epimediumhydrolysis temperature,pH value of examining factors,to examine indicators of protein removal and polysaccharide loss rate,response surface methodlogy to predict the optimum conditions.The results showed that 5%-7%in the amount of enzyme,reaction temperature50-55℃,pH5.5-6.0 conditions,protein removal was 57.22%,polysacchardes loss rate of 24.11%.Concluded through experiments design-Response Surface Optimization of the enzymatic removal of epimedium polysaccharide technology,the method is simple,good predictability.The thesis has done explorative research on analysis and research,preparation technology,purification of herbal polysaccharide incluing epimedium and achieved a significant progress.lt is wished to provide a reference for similar work in further study.
