Betula costata Trautv.is a plant of Betula L.family.Its bark has the functions of clearing heat and detoxicating,relieving cough,relieving asthma,diminishing inflammation,sterilizing,etc.,and is used for treating chronic dysentery,hematuria,acne,freckles and other diseases.Nine compounds(marked as SH-1~SH-9)were isolated and identified from 85% ethanol extract of the bark of Betula costata.According to the data analysis of NMR and IR,the nine compounds were elucidated as Betulin(SH-1),Lupeol(SH-2),3-O-Acetyloleanolic acid(SH-3),Betulonic acid(SH-4)?Betulone(SH-5)?17?-Hydroxy-28-nor-18?(H)-lup-20(29)-en-3-one(SH-6),Amygdalin(SH-7),Acerogenin E(SH-8)and 16-Hydroxy-17-O-methylacerogenin E(SH-9).Among which,17?-Hydroxy-28-nor-18?(H)-lup-20(29)-en-3-one was isolated from natural products for the first time,16-Hydroxy-17-O-methylacerogenin E and Amygdalin were first gained from Betula costata Trautv.Based on the chemical constituents,this paper has also established a HPLC method to determin the contents of lupeol(SH-2)and 3-O-acetyloleanolic acid(SH-3).Firstly,the factors such as extraction solvent,extraction method,extraction time and ratio of liquid to material were investigated before the best extraction method had been established.Secondly,the methodology was investigated.Experimental results showed that compound SH-2 and SH-3 gave a good linear relationship between peak area(Y)and the contents(X)in the range of 0.08-2.50 mg/mL and 0.16-5.00 mg/mL,respectively.Both compounds had a good performance in precision,stability,repeatability and recovery with their RSDs less than 2%.It shows that the method has good precision and high accuracy.In the results of contents determination,The contents of SH-2 and SH-3in the bark of Betula costata Trautv from Xinbin county are 1.83 mg/g and 5.25 mg/g repectively,and the contents of SH-2 and SH-3 are 0.14 mg/g and 4.66mg/g repectively for the sample from Jingyu County.In the bark of Betula platyphylla collected from Jingyu county,the contents of SH-2 and SH-3 are 1.15 mg/g and 1.25 mg/g repectively.The results showed that the contents of SH-2 and SH-3 were ont only different among the same plants,but also between the contents of SH-2 and SH-3.In summary,based on the research of scholars at home and abroad,in this paper,9 compounds were isolated and identified from ethanol extract of the bark of Betula costata Trautv,two compounds were isolated from the plant for the first time,and one was isolated from natural products for the first time.Furthermore,this paper also established the contents determination of two kinds of Triterpenoids by HPLC-DAD method.The research results of this paper provided a new scientific basis for the establishment of quality standard and the development and utilization of Betula costata Trautv.
