This study is a part of “key technology research and industrialization of corn special formula oil production” in Jilin Province,one of the core technical projects of “double tenth engineering”.Optimization of preparation and formulation was accomplished by the entrapment efficiency of folate liposomes.The liposome was made by using film dispersion-ultrasonic emulsify method,with HSPC,cholesterol and DSPE-PEG2000 as materials,and the entrapment efficiency was measured by RP-HPLC.Optimized the proportion of materials based on the response surface analysis.This experiment determined the fatty acid in oils by GC-MS technique and obtained the best blending process by orthogonal analysis.The main results ware as follows:Folic acid was stable at room temperature,light-avoiding and neutral condition,and the content remained at more than 90% after 24 h.The liposome was made by using film dispersion-ultrasonic emulsify method according to the characteristic of folic acid.Comparing with the liposomes which were prepared by span,tween and lecithin,the liposome prepared with lecithin had better stability and we selected lecithin for the next experiment.The chromatographic detection conditions were determined according to the chromatographic peaks: reverse C18 column,the ratio of methanol to PBS buffer was 20:80,the flow rate was 0.9m L/min,room temperature,the detection wavelength was 265 nm,detection time was 10 min,sample volume was 20μL.The encapsulation efficiency can be calculated by dialysis method and dialysis time was 14 h.With single factor test and response surface test,the result were as follow: the ratio of HSPC and cholesterol was 2.71 : 1(w/w),the ratio of DSPE-PEG2000 and embedding materials was 2:3,the folate concentration was 0.1 mo L/L,p H of PBS was 7.37.The encapsulation of folate liposome prepared under this condition was 53.59%.The content of fatty acids in blending oil was tested by GC-MS.This blending oil was composed of corn oil,sesame oil,palm oil,flax oil,peanut oil and olive oil,the ratio of the oils was 30:24:15:18:13:16(v/v)and in this case the optimal ratio of fatty acid was the best.The optimum process of blending corn oil according to peroxide value and acid value was as followed: the amount of TBHQ was 100 mg,the amount of VE was 100 mg and mixing time was 40 min.The shelf life of corn blend oil was predicted according to Arrhenius empirical formula and Schall oven method,the result was as followed: the shelf life of corn blend oil can reach 18 months and it was stable during shelf life.The corn blend oil was easy to preserve and kept high quality when compared with other vegetable oil.The results showed that the physicochemical indexes of the yoghurt matched up to the national standards of China.Folate liposomes were evenly distributed between 100~150nm and the properties were stable and well embedded.In order to obtain the content of folic acid in corn blend oil,we choose the microbiological method and the regression model was y=0.9046x+0.0245,R2=0.9927.The results showed that the concentration of folic acid in corn blend oil was about 1992.56μg/100 g,which could meet the daily needs of pregnant women and achieve the desired results.Finally,the study got a kind of functional oil and its future large-scale production has laid a good foundation.
