Compared with the small molecules,Polymeric assemblies own high drug-loading, long metabolic characteristics, these advantages make them become the hot spots of a diagnosis and cancer research. Because of the cell surface natural barrier and the carrier itself, The therapeutic efficiency of nano-carriers with a random helical structure on the surface is always low,In order to improve this shortcoming, we synthesized a hydrogen peroxide and acid-responsive amphiphilic polymer PLLA(-IR780)-HBPPI-HPPPI. In a neutral aqueous solution, the copolymer can be assembled into onion-like spherical micelles with uniform particle size (about 100 nm),Because it contains a single left-handed helix-formed polymer micelle that gives it a higher cell penetration efficiency,which is similar to cell-penetrating peptides. The isocyanide polymer of phenylboronic acid pinacol ester forms a dense,hydrophobic shell in the middle, the neutral or weak acid aqueous solution, shell is relatively stable.Because of this stable structure, it can reduce drug leakage and reduce the toxicity of the system cells. The ester bonds on this polymer turn back into carboxyl groups,which make a transition from hydrophobic to hydrophilic,so that the shell dissociates. This process of transformation is accompanied by the release of the drug and the production of two hydrophilic channels.In the acidic environment, the hydrophobic core of PLLA is further hydrolyzed, and the innermost drug is also released?Due to the micelle-encapsulated IR780, this carrier has the function of hyperthermia, which can rapidly convert light energy into heat energy at 808 nm (the power of the laser used is 1 W/cm2), and the temperature is raised to 50? quickly,Temperature is enough to kill cancer cells.We combined this hyperthermia and chemotherapy together and demonstrated that the treatment was significant in the trial. The focus of this study is to develop a rapid drug delivery system that can produce sustained drug release in cells. The preparation of this versatile micelle has great potential application in disease diagnosis and treatment.
