Breast feeding receives much more attention with the improvement of living standards.There is no safe and effective western medicine to treat milk-production deficiency.TCM are broadly used in clinical treatment of postpartum milk-production deficiency.However,effectiveness of most TCM cannot be confirmed due to lack of preclinical and clinical data.As a result,development of a galactagogue TCM with well-defined efficiency and safety has broad market prospect.First of all,according to compositional rules and regulatory requirement of galactagogue TCM,three compositions were obtained,respectively,group a,group b,and group c.Secondly,extraction process was studied and totally 6 feasible extraction processes were obtained: group a extraction process 2(A2)is alcohol extraction-water precipitation of Angelica sinensi,together with water extraction-alcohol precipitation of Radix Rehmanniae Praeparata,Astragalus mongholicus,Vaccaria segetalis,pig nail,and Liquidambar formosana Hance;group a extraction process 3(A3)is saponin and polysaccharide components extracted from Astragalus mongholicus,phenolic acid components extracted from Angelica sinensi,together with water extraction-alcohol precipitation of remaining herbs;group b extraction process 2(B2)is alcohol extraction of for Ligusticum chuanxiong,baicalin extracted from Astragalus mongholicus,together with water extraction-alcohol precipitation of remaining herbs;group b extraction process 3(B3)is baicalin extracted from Astragalus mongholicus,saponin extracted from Ligusticum chuanxiong and Paeonia lactiflora,together with water extraction-alcohol precipitation of remaining herbs;group c extraction process1(C1)is water extraction-alcohol precipitation of all herbs;group c extraction process2(C2)is alcohol extraction of Angelica sinensi,and dry ginger,together with water extraction-alcohol precipitation of remaining herbs.Extraction rates of 6 processes are0.206,0.213,0.158,0.132,0.289,and 0.313.Thirdly,postpartum milk deficiency rat model and healthy lactating rat model were used to evaluate the effect of TCM compositions and extraction processes on milk production.It suggests that group c extraction process 2(C2)could significantly improve single-time milk production and total milk yield comparing to model group and other treatment groups,which is close to healthy lactating group.C2 group also shows increased concentration of serumprolactin,as well as hyperplasia of mammary gland bubbles and tubes in lactating rats.These results indicate C2 has better galactagogue effect comparing to other compositions and extraction processes,which shows no significant difference in single-time milk production,total milk yield,as well as serum level of prolactin,5-HT,and cortisol.Thus group c is the optimized galactagogue composition,which contains Angelica sinensi(monarch)10g,walnuts(assistant)4g,processed dried Ginger(assistant)2g,Astragalus mongholicus(minister)10g,Platycodon grandiflorum(guide)3g,Radix glycyrrhizae(assistant)2g,prescription totals 31 g.Process C2 is the optimized extraction process: alcohol extraction of Angelica sinensi,and processed dried Ginger;water extraction-alcohol precipitation of walnuts,Astragalus mongholicus,Platycodon grandiflorum,and Radix glycyrrhizae.The mechanism of TCM C2-stimulated milk production in levodopa-induced milk deficiency rat model may involve promotion of prolactin secretion,inhibition of dopamine secretion in hypothalamus,improving hyperplasia of mammary gland bubbles and tubes,which makes C2 a good candidate for galactagogue TCM with great market prospects.
