The objective of this study is to investigate the effect of arginine on the stability of hemoglobin and the effect of arginine-hemoglobin on the color stability of ground lamb during storage.Arginine was chosen to improve the stability of hemoglobin by screening.The optimum conditions of the preparation process of arginine-hemoglobin were optimized by response surface method.The second structure and stability of arginine-hemoglobin were analyzed by circular dichroism spectrum and ultraviolet spectrophotometry,finally,this research studies the effect of arginine-hemoglobin on the color stability of ground lamb during storage.The detailed results list as follows:(1)The relative content of methemoglobin(MetHb%)and redness(a*)value were used as the index to evaluated the color stability of hemoglobin,the a* value increased significantly,and the MetHb% significantly decreased in arginine treatment group(P<0.05).Then,the single-factor experiment was designed to investigate the effect of arginine concentration,temperature,reaction time and pH on the a* value and the relative percentages of different hemoglobin derivatives(oxyhemoglobin,hemoglobin and methemoglobin).The optimum conditions of the preparation process of arginine-hemoglobin were optimized by response surface method.Results revealed that the optimal conditions for the preparation of arginine-hemoglobin were as follows: the concentration of arginine,10.51mg/mL;the pH of hemoglobin solution,10.75 and 18°C for the reaction temperature,Under these optimized conditions,the actual content of methemoglobin was 23.36%.(2)UV scanning spectrum and circular dichroism spectra were used for analysis the change of structure of hemoglobin with different treatment,and explored the influences of different concentration of NaCl and pH on the stability of arginine-hemoglobin.The result showed that the stability of hemoglobin with different concentration of NaCl was improved in arginine treatment group.Besides,the result also indicated that the denaturation temperature of hemoglobin with different treatment were no significant difference(P<0.05).The result of particle size distribution showed that particle size of arginine treatment group was smaller compare with control and lysine treatment group.In addition,the arginine treatment group could improve a* value of hemoglobin value during storage.(3)The effect of arginine-hemoglobin on color stability of ground lamb during storage was investigated.Meat color,percentage of metmyoglobin(MetMb%),thiobarbituric acid reactive substance(TBARS)and carbonyl content were analyzed under different concentration(0,0.15,0.25,0.5 g/kg)of arginine-hemoglobin during 7 days storage at 4±0.2°C.The data showed that the arginine-hemoglobin treatments had lower MetMb%,TBARS and carbonyl group content,compared with control.The redness(a* value)of treatments was higher than that in control during storage.In addition,the redness value increased as the arginine-hemoglobin concentration increased.The results suggested that the addition of arginine-hemoglobin could be an option in inhibiting the oxidation of myoglobin and fat in meat.
