| In this paper,two novel aminopolycarboxylic acid ligands,dtpa-bis(guanine),and dtpa-bis(cytosine)were designed and synthesized by improving experimental method.Their corresponding Eu(III)complexes,Eu-dtpa-bis(guanine)(Eu-dtpa-DG)and Eu-dtpa-bis(cytosine)(Eu-dtpa-DC),were successfully prepared through heat-refluxing method.Eu-dtpa-DG and Eu-dtpa-DC were served as fluorescent probe to detect Adenine(Ad)and 6-thioguanine(6-TG),respectively.As a comparison with Ad and 6-TG,these base compounds including guanine(Gu),cytosine(Cy),hypoxanthine(Hy),xanthine(Xa),thymine(Th),uracil(Ur),uric acid(UA)and 6-mercaptopurine(6-MP)were selected as the interference substances to verify the selectivity of the two fluorescent probe(Eu-dtpa-DG and Eu-dtpa-DC).We take the Ad and 6-TG as research object combining respectively with the Eu-dtpa-DG and Eu-dtpa-DC to undertake experiment in this paper.How the Eu-dtpa-DG and Eu-dtpa-DC to detect the Ad and 6-TG is the main purpose of this paper,which have the very important significance for understanding their dosages or residue concentration in the organism of Ad and 6-TG as drugs to treat diseases such as ulcerative colitis,dermatitis,rheumatoid arthritis and biological effects.Therefore,the determination of concentration level of Ad or 6-TG is considered to be a challenging and important task in the field of physiology and clinical pathology.Base compounds are not only participate in the synthesis of genetic material playing a crucial role in life process,but also fulfiling a variety of functions in the metabolism of the cell.In recent years,Ad can be used as drugs for leukopenia caused by tumor chemotherapy disease,also used in acute granulocytopenia.Moreover,the abnormal change of Ad in organism may indicate the mutation and deficiency of the immune system leading to the manifestation of various diseases including epilepsy,cancer,tumorigenesis,mental retardation,HIV infection,Parkinson’s disease,carcinoma and liver diseases.6-TG,is a close structural analogue of Gu,was introduced into the clinic and first studied in patients in the early 1950 s.6-TG has long been used as a popular drug for leukemia treatment,similar to the 6-mercaptopurine(6-MP),in the last two decades.In recent years,it has been applied to treat breast cancer and influence immune response in the treatment of inflammatory diseases such as ulcerative colitis,dermatitis and rheumatoid arthritis.Therefore,study of base compounds has been the focus with more attention in the biomolecular research.This paper mainly contains two parts as following:1.In this section,dtpa-bis(guanine)compound and its corresponding Eu(III)complexes,Eu-dtpa-bis(guanine)was successfully prepared through heat-refluxing method.This part mainly studies the reaction process and mechanism between Eu-dtpa-bis(guanine)and adenine.By fluorescence spectroscopy,we have studied the effects of Ad concentration in the fluorescence spectrum of Eu-dtpa-bis(guanine)complex.Meanwhile,in this work,the effects and interferences of Gu,Hy,Xa and Ua were also studied.By ultraviolet-visible(UV-vis)and fluorescence spectroscopy,it can be found that the Eu-dtpa-bis(guanine)complex interact with the Ad,Gu,Hy,Xa and Ua by the different way,respectively.Through the above experiments,we can predicte and analyze the reaction process and mechanism between Eu-dtpa-bis(guanine)and adenine.2.In this section,dtpa-bis(cytosine)compound and its corresponding Eu(III)complexes,Eu-dtpa-bis(cytosine)was successfully prepared through heat-refluxing method.This part mainly studies the reaction process and mechanism between Eu-dtpa-bis(cytosine)and 6-thioguanine.By fluorescence spectroscopy,we have studied the effects of 6-TG concentration in the fluorescence spectrum of Eu-dtpa-bis(cytosine)complex.Meanwhile,in this work,the effects and interferences of Cy,Gu,Ad,Hy,Xa,Th,Ur,UA and 6-MP were also studied.Furthermore,The influence of pH on the detection of 6-TG using the EuIII-dtpa-bis(cytosine)complex as a fluorescence probe was examed.By ultraviolet-visible(UV-vis)and fluorescence spectroscopy,it can be found that the Eu-dtpa-bis(cytosine)complex interact with the Cy,Gu,Ad,Hy,Xa,Th,Ur,UA and 6-MP by the different way,respectively.Through the above experiments,we predicted and analyzed the reaction process and mechanism between Eu-dtpa-bis(cytosine)and 6-thioguanine.Besides,we have also studied the detection of 6-TG in the actual urines.It was found that the fluorescent probe could be reliable and effective for the determination of 6-TG in real urines. |
