Metal–organic frameworks(MOFs)are a class of crystallized porous polymeric materials consisting of ions or clusters linked together by organic bridging ligands.Compared to traditional porous materials,they possess predominance of large surface areas,tunable pore size and shape,adjustable composition and functionalized pore surface,which enable them unique advantages and promises for applications in gas adsorption,separation and drug delivery systems.5-Fluorouracil(5-Fu),a pyrimidine analogue with antimetabolic,is widely used in a variety of tumors.Due to the short half-life and the side effects of the drug,the development of the disease is limited.In order to overcome this shortcoming,three kinds of MOFs with different structures were synthesized,then loaded with anticancer drug 5-fluorouracil,and drug carrier and in vitro release properties were investigated.In this study,three kinds of MOFs(MIL-100(Fe),MOF-5 and ZIF-8)were synthesized by solvothermal method using Fe and Zn as the central metal,and 1,3,5-benzoic acid,terephthalic acid and 2-methyl imidazole as organic ligands,respectively.After separation and purification,the structures and properties of the synthesized MOFs were approved by SEM,XRD,TG and FTIR.The UV spectrophotometry method was firstly established to investigate the pharmaceutical preformulation studies of 5-Fu.The results showed that the mass ratio of MOF to 5-Fu and the time of action could affect the drug loading,the highest drug loading rates are 0.261g/gMOF for MIL-100(Fe),0.296g/gMOF for MOF-5 and 0.505g/g MOF for ZIF-8.The results showed that the drug encapsulation did not alter the structure of MOF.The in vitro release experiment was carried out by dynamic dialysis method.It was proved that the MOFs loaded with 5-Fu could be used to enhance the antitumor effect of 5-Fu and the drug concentration in the lesion site.It can be used as a targeting drug delivery system,which can reduce the toxicity of 5-Fu and improve the bioavailability.
