Marine microorganism is a novel kind of natural resources and it has large reserves,wide usage and renewability,it has become a new resource.It is famous in the fields such as natural green food and diesel oil.At present,biological effects of marine microorganisms has attracted lots of attention.This article has taken deeply research in Schizochytrium,and found activities of its enzymatic hydrolysis product.Also peptide sequences of the effective component have been predicted.The material in this article was originated from Schizochytrium dross.The dross was enzymatic hydrolyzed by protease,and then be used in mice experiment in vivo and cell culture in vitro to,in order to test its effect.In vivo,we mainly test the anti-inflammatory effect of product.This experiment was based on DSS-induced acute enteritis model on C57BL/6 mice.We used enzymatic hydrolysis product as experience group,PBS as negative control group and Norfloxacin as positive control.We recorded body weight,stool,intestinal condition,intestinal mucosal injury and inflammatory cytokines of each group.By comparison,we found that the product has a significant effect on treating intestinal inflammation,even better than Norfloxacin.Then,obesity model of KM mice was established to test the obesity inhibition effect of the product,but the result was not significant.In order to find more effect of the product,this article choose 293T cell as research object.We added different concentration gradient of enzymatic hydrolysis product into cell culture medium.Cell growth status and cell survival rate at 24 h,48 h and 72 h were observed and recorded.The result showed that concentration of enzymatic hydrolysis product at 0.1 mg/mL could significantly increase the proliferation and survival rate of 293T cells.This indicated that the enzymatic hydrolysis product of Schizochytrium dross had the effect of promoting cell growth.By comparing the effect of dross and product,we found that the efficacy of anti-inflammation was originated from the components after enzymatic hydrolysis.Analyzed by Q-TOF-MS/MS,32 possible molecular weight components were found.By compared with PeptideCutter result of the 10 currently known protein sequences of Schizochytrium from NCBI,169 possible peptide sequences were obtained.The follow work of our group is to select the activity peptide sequences and synthesis in the future.We hope this product could have a high social value as a kind of functional food or medicine,thus turn the waste to treasure.But due to its raw material is origin from the sea,the strong fishy smell will limit its application.We have used activated carbon adsorption method to remove this bad smell and the result was very effective.After analyzed the components before and after treatment by UPLC-MS,we found that the components had almost no changes after carbon adsorption.Then micro capsule technology was used to beautify the product's appearance and extend its shelf life.
