| In this study,soybean powder was hydrolyzed in n-hexane by lecitase ultra to prepare phospholipids acid.Firstly,the effects of reaction time,temperature,enzyme dosage,substrate concentration,water content and p H value on the reaction rate were investigated.Then,the optimal reaction conditions(reaction time,12.1h,temperature,45℃,enzyme dosage,3%(w/v),substrate concentration,20.8%,water content,23.8%,and pH,6,were obtained by response surface methodology.Accordingly,the acid value of phospholipids lysophosphatidic acid in the optimal reaction condition was 76.7 mgKOH/g.In addition,the relative contents of phosphatidyl choline,1-phosphor-lipin acid radical choline and 2-acyl-were lysophospholipids acyl choline were determined as 15.51%,3.99% and 19.6%,respectively.Using soy lecithin concentrates as raw material,under certain conditions,by supercritical CO2 hydrolysis method by the hydrolysis of phospholipids,extraction of oil were lysophospholipids get off.Test influence supercritical CO2 extraction after the main factors of phospholipids hydrolysis temperature,pressure,time,and investigated through orthogonal experiment of process conditions are optimized.The experimental results show that 54℃,extraction extracting temperature 29 MPa,extraction time 5.8h pressure for the best conditions,doffing were lysophospholipids extraction rate reaches 92.4%.Column chromatography method was used for separate and purify the crude product of the lysophosphatidic,(Chromatography)silica gel was used as adsorbent,eluent,flow rate and sample size and other factors were investigated;According to experimental data to determine the optimum conditions were: chloroform: methanol 2:1 mixed system as the eluent,flow rate 3ml/min,plus sample(sample mass / adsorbent mass)0.025 lysophospholipids(gsilica gel)-1,used this conditions:the lysophosphatidic the purity of 96.9%,the recovery was 93.6%。Phospholipid hydrolysis of lysophospholipids crude product as a raw material,after the acetone elution processing,under certain conditions research in the elution temperature,elution time,eluent ratio of elution times on the effect of purification of lysophospholipids;get the optimum conditions were: elution time 15 min,elution temperature of 0 ℃,liquid feed ratio of 15:1,elution times 8 times.The lysophosphatidic get in this condition,the purity of 93.2%.Selecting lipase Novozyme 435 as a tool enzyme,response surface method for the numerical analysis,lysophosphatidic transesterification tests in solvent-free vacuum system.The response surface methodology and regression analysis to determine the optimum conditions were: reaction time was 3.04 h,the enzyme concentration was 2.36%,Moore bottom material ratio of 4.43,the water activity of 8.44%,the reaction temperature is 50 ℃.Under these conditions,the hemolytic phosphate exchange binding rate of up to 25.4. |
