| In order to substantially increase the added value of rapeseed processing,and ease the tight market demand of phospholipid products at all levels simultaneously,this experiment improved the production processes of de-oiling and liquidity improving of rapeseed phospholipid.Then improved the purity of lecithin by using the method of organic solvent liquid-liquid extraction to the food grade phospholipid and pharmaceutical grade lecithin standards.More over,it used the metal ion sequestration resin adsorption method to purify lecithin.The main results were as follows:First,the most economic conditions of oil off by acetone extracting was investigated.Solid-liquid ratio was 1:1.The result of liquidity improving experiment concluded that the impurity content was≤ 2%,phospholipid concentration was ≤ 60%were the best condition.And it obtained that solvent residue met the food grade standards of phospholipid by gas chromatography.Secondly,on the basis of crude lecithin obtained by single ethanol extraction,purification technology of organic solvent liquid-liquid extraction was studied:liquid-solid ratio was 6.48:1,liquid water content was 0.06,ratio of solvent was methanol:hexane = 1:0.93,the extraction temperature was 45 ℃,shaking time of constant temperature water bath was 40min.Under these conditions the purity of lecithin was 72.6%and the recovery was 42.6%.Then static exchange metal ions with various resins was studied.Determined the D113 resin as non-functional group carrier to fix metal ion,and copper ion as the best metal ion.Through the study of lecithin loading conditions of column chromatography,the result have been concluded as follow:loading amount was 4.50g·(L resin)-1;flow rate was 0.43mL/min;loading concentration was 5.00g/L.Under these conditions the purity of lecithin was 80.2%and the recovery was 68.2%.All levels of lecithin treated by the methods above met domestic and international soybean phospholipid standards.Can be widely used in food,pharmaceutical industries improve the added value of rapeseed process.If industrialized,it will reduce the demand of domestic lecithin import. |
