Monoraphidium dybowskii C29 belongs to Chlorophyta,Monoraphidium.Monoraphidium dybowskii C29 has many advantages such as rapid growth,high biomass,high oil content and so on.The Moonoraphidium dybowskii C29 was chosen as research material to explore its large-scale cultivation,nutrient components,chemical components and capacity of sewage treatment.The results were as follows:The effects of temperature,light intensity,nitrogen sources,feeding strategy and inoculum concentration on the growth of Monoraphidium dybowskii C29 were investigated,which showed that Monoraphidium dybowskii C29 could adapt itself to the temperature ranging from 15 to 30 ? and the outdoor light intensity could also meet the demands of its growth.In addition,urea was chosen as the nitrogen source because of its lower price and there was no significant difference between the groups using urea and those using other nitrogen sources.Besides,the growth of Monoraphidium dybowskii C29 when fed in batches was superior to that when fed once and the way of feeding in batches was propitious to pipelines' cleaning.The increase of inoculum concentration was proved to be good for the growth of Monoraphidium dybowskii C29.Under the controlled conditions,the biomass of Monoraphidium dybowskii C29 was 1.15 g/L and the lipid production was 0.39 g/L when cultivated in mass outdoors.The Monoraphidium dybowski C29 was proved to be highly adaptive to the environment,which could be achieved cultivate in a larger scale.As for the nutrient components,the results showed that Monoraphidium dybowskii C29 was rich in grease(30.25%),fiber(23.77%),protein(20.08%),carbohydrates(11.31%)and pigment(4.22%).In addition,the contents of magnesium,calcium,zinc and ferrum were higher than other mineral elements;the contents of heavy metal elements such as Pb,Cr,Cd,As,Ni also met the requirements of national standards;there were 18 kinds of amino acid in Monoraphidium dybowskii C29,taking up 17.68%and the content of essential amino acid accounted for 37.44%of total amino acid,which proved to be high in essential amino acid index(EAAI).According to essential amino acid index(EAAI),the protein in Monoraphidium dybowskii C29 is of high quality.Besides,the contents of VB6(51.26 mg/100 g algae powder)and VC(19.88 mg/100 g algae powder)were the highest among 8 kinds of vitamins and the contents of linoleic(25.86%)and linolenic(9.27%)among fatty acids were higher.There are rich nutrient components in the Monoraphidium dybowskii C29,which can be used as the raw material of food and feed.As for chemical components,the different components of Monoraphidium dybowskii C29 with anti-bacterial and anti-oxidative activities were investigated with oxford plate assay system,DPPH radical-scavenging and FRAP,from which we found that both anti-bacterial and anti-oxidative activities of the ethyl acetate extracts(EtOAc extracts)were stronger than the mineral ether,n-butyl alcohol and water extracts.The EtOAc extracts were selected for further separation.Accordingly,three liposoluble constituents were obtained.There were 48 chemical components were isolated and then,identified,which could be mainly divided into esters,alkyl acids,alkyl sioxanes and alkanes.Then,the anti-bacterial and anti-oxidant experiments were conducted on the components eluted from EtOAc extracts by different polarity,from which two(mineral ester:EtOAc=90:10 and 80:20)high in anti-bacteria and anti-oxidation activities were screened.Eventually,four compounds were isolated from the two extracts by silica gel chromatographic method and high performance liquid chromatographic method(HPLC)and identified on the basis of Nuclear Magnetic Resonance(NMR),which proved to be Indole-3-carboxylic acid,(+)-Epiloliolide,(-)-Loliolide and 5-Hydroxy-3,4-dimethy-5-pentyl-2(5H)-furanone.These four compounds were isolated from Monoraphidium dybowskii C29 for the first time which had anti-bacteria and anti-oxidant activities,and the compound 1 showed the best anti-oxidative activity.On this part,the capacity of Monoraphidium dybowskii C29 on sewage treatement was studied.The nutrient components in domestic sewage could not meet the demands of the growth of Monoraphidium dybowskii C29.Different concentration of BG11 media(no nitrogen and phosphorus)were added into domestic sewage,and when BG11 media(50%)were added,the growth of Monoraphidium dybowskii C29 was best and the removal rates of nitrogen and phosphorus reached the maximum,which were 86.09%and 90.64%,respectively.Simultaneously,the effect of Monoraphidium dybowskii C29 on domestic sewage after diluted was also investigated.When the volumn fraction of domestic sewage was 60%,the growth of Monoraphidium dybowskii C29 was best and the removal rates of nitrogen and phosphorus reached the maximum,which were 93.31%and 96.51%,respectively.In addition,the removal rates of eight kinds of metal ions in domestic water were over 50%and the removal rates of Ni,Cu,Cd and Pb were over 60%.
