| Perilla is a traditional Chinese herbal medicine and oil crops.There contain more oils in perilla seeds and procyanidins in its skins,so it has a glorious prospect to develop comprehensively perilla seeds.In this study,the oil was obtained by the ultrasound-assisted aqueous enzymatic from perilla seeds,the optimal conditions were investigated,and its characteres were analyzed.At the same time,the technologies of extraction and purification of procyanidins in perilla seed skins were also studied,and its antioxidant activities were analyzed.The main results as follows:(1)Based on the single factor tests,the ultrasound-assisted aqueous enzymatic extraction technique of perilla seed oil was optimized by response surface methodology.The optimal process parameters were enzyme proportion was cellulase 5.5%,neutral protease 4.5% and pectinase 7.5%,liquid-solid ratio 6:1,ultrasonic time 23 min,enzyme hydrolysis temperature 50℃,enzyme hydrolysis time 2.5h,and the yield of oil was 30.45%.(2)The physicochemical properties,fatty acid composition,the content of bioactive components and the antioxidant activities of perilla seed oil extracted by ultrasound-assisted aqueous enzymatic(UAAE)were evaluated and compared with those of oil obtained by hot pressing extraction(HPE),cold pressing extraction(CPE)and solvent extraction(SE).The results indicated that UAAE oil had a lower peroxide value and a higher iodine value.The UAAE oil had a lower palmitic acid content,the oleic acid content of UAAE oil is significantly higher than that of HPE oil or SE oil,and UAAE oil contained higher levels of α-linolenic acid.The VE content of UAAE oil is not much different from that of SE oil.The UAAE oil had the highest content of polyphenol,total flavonoid and sterol.The UAAE oil also had superior antioxidant activities by DPPH,ABTS and FRAP assay.On the whole,the quality of UAAE oil is well.(3)The ultrasound-assisted extraction technique of perilla seed skin procyanidins was optimized by single factor tests and response surface methodology.The optimal process parameters were liquid-solid ratio 15:1,ethanol volume fraction 62%,ultrasonic time 30 min,ultrasonic temperature 53℃,and the yield of procyanidins was 2.170mg/g.(4)The optimal macroporous resin was obtained by static adsorption and desorption tests,which the best resin was XDA-8.The optimal dynamic process parameters were obtained by dynamic adsorption and desorption tests,which included the sample containing 4mg/m L,flow rate for sample of 4BV/h,elution with 70% aqueous ethanol at a flow rate of 2BV/h,and its amount for elution of 13 BV.Under these conditions,the content of perilla seed skin procyanidins was increased from 5.25% to 12.10%,increased by 130.48%.The antioxidant capacity of four kinds of Perilla seed skin procyanidins before purification,purified procyanidins,VC and BHT were measured and compared,it was found that the purified perilla seed skin procyanidins had the strongest antioxidant ability. |
