| The oat bran polysaccharide exhibited many physiological functions according to the researches,such as balance the cholesterol level,decreasing blood pressure,hypolipidemic effect and enhancing the immunity.On basis of our laboratory previous research,this study focus on the improvement of the oat bran polysaccharide yield.The blood lipid regulation of the oat bran polysaccharide for the hyperlipemia model rat was also investigated.The research of the ultrasonic microwave extraction.The optimal condition of extracted polysaccharide from oat bran was investigated with the ultrasonic-microwave synergistic method in order to improve the yield.The effects of four factors,microwave power,liquid to solid ratio,p H,ultrasonic-microwave synergistic time,on the yield and purity of oat bran polysaccharides were studied.The optimal extract conditions of the polysaccharides were as follows,microwave power 639 W,the liquid-to-material ratio of 36:1,the p H 10 and extraction time 18 min,which was obtained by the two regression center combination test and the response surface method.The yield increased from 4.30% to 8.45% compared with the traditional water extraction method,and the structure of the polysaccharides also didn’t show any change according to the result of scanning electron microscope.The research of the establishment of hyperlipidemia rat model.Two ways of establishing hyperlipidemia model in SD rats with fat emulsion and high-fat feed were investigated in this part in order to establish a highly efficient and stable hyperlipidemia animal models,as the index of rats weight,six blood lipid level,and pathological section.As a result,there were significant difference of the six blood lipid level between the high fat feedstuff group,high-fat emulsion rats and blank group after 4 weeks.After 5 weeks,the stability of the high-fat feed group model is higher than the high fat emulsion group.There were significantly difference between the blank group and high-fat feed group for the fat droplets size,number of pathological on the basis of liver pathology section.The study of antilipidemic effect of the oat bran polysaccharide on the hyperlipidemia rats.After the establishment of hyperlipidemia animal models,the rats were fed with oat bran polysaccharide(high,medium and low doses),oat bran powder(high,medium and low doses)and beta glucan health products sold(high,medium and low doses)in addition to the blank control group and high-fat model control group.Various indexes were determined to evaluate the oat bran polysaccharide on the hyperlipidemia rats.The medium dose group of oat bran polysaccharide,high dose group of oat bran powder,high dose group of beta glucan exhibited the best antilipidemic effect.Compared with the blank group,the TC,HDL-C and LDL-C decreased 1.079 mmol/m L,0.579 mg/d L and 0.489 mg/d L,respectively,and TG increased 0.341 mmol/m L for the medium dose group of oat bran polysaccharide treatment.The TC,HDL-C and LDL-C decreased 0.804 mmol/m L,0.586 mg/d L and 0.529 mg/d L,respectively,and TG increased 0.286 mmol/m L for the high dose group of oat bran powder treatment.The TC,HDL-C and LDL-C decreased 1.104 mmol/m L,0.799 mg/d L and 0.548 mg/d L,respectively,and TG increased 0.227 mmol/m L for the high dose group of beta glucan treatment.The improvement of the adipose hollow space was the best for the oat bran polysaccharide in medium dose group,oat bran powder in high dose group and beta glucan in high dose group,according to the liver pathological section.In conclusion,the oat bran polysaccharide play a regulatory role on the blood lipid level of hyperlipidemia rats.The research lays the foundation for the further development of oat bran polysaccharide as the healthcare products. |
