Background: With the rapid and healthy development of the market economy and its reform,the variety of food has increased and some health foods have appeared.Health food is a type of food,which has the general commonality of food.In the meanwhile,it can regulate the body's various function,to adapt to specific groups of people.In recent years,accompanyed by the rapid development of food industry,food safety scandal surfaced,fake and shoddy foods abound,food additives accounted for a great proportion.The incident caused a public panic which contains taurine ingredients.Taurine as a food additive,appeared in a variety of health food and medicine.The accuracy of the amount content of taurine on the label of health food and medicine has been become a significant issue.It will have an impact on the normal people.Food safety incidents have attracted people's attention,it is necessary to detect and research for health food analysis.Objective:Due to the pretreatment method of taurine is complexed,according to the characteristics of taurine,using different solvent extraction,simiplfied the pretreatment process.Establishing two method for the determination of taurine content in health food.Namely,thehigh performance liquid chromatography-evaporative light scattering detector and high performance liquid chromatography-tandem mass spectrometry.These two methods can be used for the detection of similar health foods.Method:The pretreatment was optimized from extraction solvent,mobile phase'type and concentration.To establish a detection method of taurine in health food application of ultra-performance liquid chromatography-evaporative light scattering detector(UPLC-ESLD)and a detection method of taurine in health food application of Liquid chromatography--mass spectrometry.The accuracy and reliability of the two methods are verified by precision,recovery and stability tests.Result: 1.Pre-treatment process: acetonitrile-water(1:9,v:v)as extraction agent.According to different types of samples,using different ways to deal with it.The capsule was dissolved by adding chloroform.supernatant was filterd by the water-membrane.The average recovery is above80%.The Waters BEH Amide(50mm×2.1 mm,1.7?m)chromatographic column,The mobile phase was eluted with0.1% formic acid and acetonitrile water,it can effectively separate the taurine of 13 Kinds of health food.There is a good linear relationship in range of 0.10mg/m L~2.0mg/m L,the linear regression coefficient R2 were 0.9999.Standard addition recovery range of 98.5%~100.7%,the relative standard deviation ranged from 0.48% to 1.29%,the quantitative limit of Methods was 0.1mg/m L.This method shortens the taurine of13 kinds of health food detection time and improve the detection efficiency.2.Pretreatment method: acetonitrile-water(1:9,v:v)as extraction solvent.According to different types of samples,using different ways to deal with;The capsule was dissolved by adding chloroform.The supernatant was filtered by the water-membrane.The average recovery is above 80%.The Waters BEH Amide(50mm×2.1mm,1.7?m)chromatographic column.The mobile phase was eluted with acetonitrile-water,it can effectively separate the taurine of 12 kinds of health food.There is a good linear relationship in range of 0.01?g/m L~2.0?g/m L,the linear regression coefficient R2 were 0.9994.Standard addition recovery range of 97.5%~100.8%,he relative standard deviation ranged from 0.67% to 3.9%,the quantitative limit of the methods was 1.0mg/kg.This method shortens the taurine of 12 kinds of health food test time and improve the accuracy of the results.Conclusion:1.It established a method for determination of taurine by ultra high performance liquid chromatography coupled with evaporative light scattering and liquid chromatography tandem mass spectrometry.The utility model has the advantages of simple operation,high efficiency and sensitivity,and meets the current national testing standards the requirements of daily similar food testing.providing technical support for the detection of working in the future.2.Compared with the national standard method,the pretreatment method of taurine content in the health-care food is simplified,the derivative method in the national standard is avoided,the cost is saved,and the utility model is simple,safe and environmental friendly.3.There are many advantages of the two kinds of detection methods,high sensitivity and precision,low detection limit and high detection efficiency,to provide technical support for the detection of taurine.
