In recent years,Chaohu blue algae eruption frequently,blooming fresh blue algae resource utilization more and more attention,especially in aquatic cyanobacteria phycocyanin extraction and purification.Studies have shown that high purity phycocyanin has a good effect,such as anti-inflammatory,anti-cancer,antioxidant and so on.But aquatic cyanobacteria in the algal toxins great harm,it is a liver toxin,with a strong role in promoting cancer(liver cancer).In this study,we obtained the crude extract of phycocyanin from the fresh algae cyanobacteria in Chaohu Lake,and obtained the phycocyanin solution by two-step salting-out method to optimize the solid phase extraction-high performance liquid chromatography The removal efficiency of algal toxins in each stage was studied by ultrasonic method and coagulation-ultrasonic combination method.The preservation conditions of phycocyanin solution after two-step salting-out were also studied.The main contents are summarized as follows:1.Determination of the types and proportions of mobile phase,detection of wavelength,column temperature flow rate and other conditions and required for the determination of algal toxin content by high performance liquid chromatography.And then on this basis,the optimal extraction of solid-phase extraction of cyanobacterial toxins in the process of eluent and eluent concentration.And finally determine the mobile phase of the detection of algal toxin:Methanol: 0.01 M ammonium acetate = 55:45;wavelength: 238 nm;column temperature: 30C;flow rate: 1 ml / min.The optimal eluent concentration for solid phase extraction was: 20% methanol solution;concentration of eluent: 100%methanol solution.2.Study on removal effect of algal tox by ultrasonic method.Based on the determination of algal toxin content by solid phase extraction and high performance liquid chromatography(HPLC),the content of algal toxins at all stages of phycocyanin solution was detected by two-step salting-out process.Results showes that most of the algal toxins were found in crude extract and two steps of salting out the supernatant.Firstly,for crude extract and two steps of salting out the supernatant,we studied the effects of ultrasonic power,ultrasonic time,solution p H on the removal of algaltoxins,then we compared with the coagulation process,ultrasonic process and ultrasonic coagulation process,The results showed that the coagulation and ultrasonic combination process had the best effect on the removal of algal toxins and the removal rate of algal toxin was the highest when the ultrasonic power was 950 W,the ultrasonic time was 10 min,the solution was neutral and the amount of coagulant aluminum sulfate was 8mg / l.3.Study on the stability of cyanobacteria from.The effects of temperature,p H value,freeze-thaw times and additives on the stability of phycocyanin solution after two-step salting-out were studied.The optimal conditions for the preservation of phycocyanin solution were determined.The results showed that the preservation of phycocyanin solution was favorable at low temperature and neutral condition.The addition of carbohydrate,preservative,oxidant and metal ion could also affect the stability of phycocyanin.
