| Giant salamander is a rare precious animal in China,and its skin can produce mucus easily.At present,the research is mainly focused on the ingredient and biological activity of mucus.In this study,the glycoprotein named MGP-III was extracted from Giant salamander mucus.The purification and purity identification,the physical and chemical properties,the oxidation resistance in vitro and the preparation process of moisturizers about MGP-III were all studied.The main methods and the results were as follows:After the freeze drying processing of Giant salamander’s mucus,the main ingredients were crude protein 82.12%,total sugar 11.71%,crude fat 4.05%,ash content 1.63%.The Giant salamander crude glycoprotein were obtained by using the method of hot water extraction and ethanol precipitation.The optimum conditions for MGP-I were obtained by the orthogonal test.The optimum conditions were:1:35,12 h,90 ℃.In this experimental condition,the ratio of obtaining crude glycoprotein named MGP-I was 8.36%.For MGP-I,the total sugar content was 55.54%,protein content was 22.03%,reducing sugar content was 10.38%.The glycoprotein MGP-III was obtained by using two purification methods of the DEAE-52 column chromatography and Sephadex G-100 column chromatography.It was to proved that MGP-III was only one ingredient by using ultraviolet absorption spectrum analytical method,SDS-PAGE electrophoresis analytical method,and HPLC analytical method.It had obvious absorption peak at 193 nm and 284 nm by UV analysis;It was only one single dyeing belt,and the molecular weight was about 58,000 Da;By using the RID test and UV test,it showed only one single and symmetry peak,and had the same retention time about 12 min of the peak.The physical and chemical analysis of MGP-III showed that the total sugar content was 61.83%and protein content was 30.32%.Through the ninhydrin,Fiji,biuret reaction,it explained that MGP-Ⅲ was one glycoprotein;After the results of hygroscopicity and moisture experiment,MGP-Ⅲ under the condition of RH 43%was higher than RH 81%,and also was higher than RH 81%on the moisturizing effect,and MGP-III had a certain moisture absorption and moisturizing effect.The MGP-Ⅲ’s reducing power,hydroxyl radical,DPPH free radical,superoxide anion radicals scavenging activities and UV resistance were detected in vitro chemical simulation system.When MGP-Ⅲ’s concentration was 1.0 mg/mL,the hydroxyl free radical clearance was 52.26%,DPPH free radical clearance rate was 61.45%,superoxide anion radical clearance rate was 50.26%respectively,and It was to be proved that MGP-III had a good free radicals and antioxidant capacity;The UVA and UVB ultraviolet transmissivity of MGP-Ⅲ was in a lower value,that illustrating MGP-III was a compound on the point of resisting the UVA and UVB rays.Research on the preparation of glycoprotein moisturizers.After the process optimization,the basic formula was that glycerol 10 g,propylene glycol 5 g,dimethyl silicone oil 3 g,glycerol monostearate 6 g,octadecanol 4.4 g,twain-20 2.5 g and MGP-III 0.002 g,fatty acid 8 g,triethanolamine 1 g,deionized water 60.1 g. |
