| In this study,Lachnum YM156 was used to obtain mycelia by deep fermentation,and the melanin was extracted from mycelia and purified to obtain intracellular melanin(LIM).The possible chemical structures of LIM were deduced by elemental analysis,UV-Vis absorption spectroscopy,infrared spectroscopy(IR),mass spectrometry(MS)and nuclear magnetic resonance spectroscopy(NMR).MTT cytotoxicity assay showed that LIM was non-toxic.Arginine melanin derivative(ALIM)was prepared.The effects of LIM and ALIM on radiation-induced injury were studied.The results showed that compared with the model group,the activities of antioxidant enzymes in the mice skin of LIM and ALIM groups were significantly increased and the content of malondialdehyde was decreased.In addition,interleukin(IL)-1α,interleukin(IL)-1β,interleukin(IL)-6,and tumor necrosis factor-α(TNF-α)were decreased.The protection properties of UVB injury of ALIM was more obvious than that of LIM at the same dose.We believe that LIM and ALIM can be used as protective agents for UV radiation damage.Chronic renal failure(CRF)was induced in mice.The protective effects of LIM,Haikunshenxi Capsule(HC)and LIM + HC on CRF mice were studied preliminarily.The results showed that LIM and HC treatment alone;LIM+HC combination could increased the levels of superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-PX),glutathione reductase(GSH)and reduced malonaldehyde(MDA)concentrations in the nephridial tissues,also reduced nephridial levels of TNF-α,IL-1β,IL-6,and activities of inducible nitric oxide synthase(iNOS).Interestingly,HC and LIM combination produced higher therapeutic effect than HC alone.This means that LIM can be applied to CRF drug research. |
