Fermentation of high concentration ethanol has important significance for the fuel ethanol industry,but high concentration of substrate and alcohol has a strong inhibitory effect on the growth and fermentation performance of yeast,so the high sugar tolerance and ethanol tolerant strains and optimization of process parameters was determined in industry product.By using the ZU108 yeast from Prof.Wu Xue-chang of the Zhejiang University,excellent strains resistant to high concentrations of ethanol,through the test of multiple batches of comparison to determine the goodness of the strain and the adaptability to the company's production of raw materials;according to the characteristics of the strain,through single factor experiment and response surface method of fermentation temperature,inoculation amount and amount of urea optimization of raw cassava,optimum fermentation conditions:temperature 33.5,inoculation amount was 25%,urea was 0.4‰;The mixture optimum fermentation conditions were as follows:the temperature was 33,the inoculation amount was 25%,and the addition of urea was about 0.3‰.Glycerol is important for osmoregulation and maintain of cellular redox balance in yeast cells.Using HPLC method,the amount of glycerol in fermentation byproduct of different strains was compared with cassava as raw materials,ZT12,ZU93,ZU108 and Angel yeast activation main by-product amount of the difference was not significant;in mixed materials,ZU108 strain decreased to 20%than Angela yeast and production strain of the yield glycerol production.By changing the process parameters that lower temperature can reduce the yield of glycerol.TO optimize the process,statistical analysis was used to analyze the test results.Using ethanol fermentation kinetic to understand the fermentation process,to demonstrate that increasing the initial sugar concentration of fermentation is beneficial to the improvement of fermentation rate.
