In this paper, passion fruit peel as crude materials for the study of dietary fiber, based on passion fruit peel component measurement on the use of response surface methodology for the extraction of dietary fiber, while using six different methods for modification extraction dietary fiber physicochemical properties and features, combined with mouse feeder experiments demonstrated and discussed some of the physiological functions of dietary fiber modified, concrete results lie as follows:(1)In this paper the determination of moisture content, ash, crude fiber, tea polyphenol, protein, fat, polysaccharides(in glucose meter), total flavonoids(in rutin meter) and vitamin B1 in the peels of Passiflora edulis indicated that the content respectively: 84.60% and 5.52%, 3.0%, 0.22% and 1.41×103(mg/kg), 1.78(g/100g), less than 0.5(g/100g),0.46(g/100g),65(mg/100g),less than 0.01(mg/100g). The results showed that the Passiflora edulis peel in rich nutrients, principal components for fiber substances, phenolic compounds, pectin and protein.(2)In this paper, passion fruit peel as crude materials for the extraction process of extraction of dietary fiber using enzyme-alkali method and enzyme-alkali ultrasonic assisted method in two ways.Optimized by single factor and response surface experiments, using multiple linear regression equation quadratic fitting the best process conditions. The best process condition of enzyme-alkali ultrasonic assisted method showed: 3% of the sodium hydroxide concentration, 1:20.5 of the radio of material to liquid, 57.83℃ of alkaline hydrolysis temperature, 0.57% of mixture of compound enzyme, dietary fiber yield of 54.93%, with a purity of 90.33%. Ultrasonic assisted enzyme alkaline optimum extraction conditions were: ultrasonic power 355.53 W, ultrasonic time 32.81 min, ultrasonic temperature was 37.32 ℃, dietary fiber yield of 53.07%, 91.02% purity. Visible ultrasonic assisted enzyme alkaline than the enzyme alkaline, the yield of dietary fiber has declined, but the purity of a slight increase.(3)The dietary fiber of Passion Fruit which was extracted by enzyme-alkali method was taken as the raw material. Modified treatment was conducted on such material with superfine grinding by dry processing, superfine grinding by wet processing, hyper-pressure method, steam explosion, enzyme method, and chemical method. In addition, study was conducted on the physical and chemical property, structure and function before and after modification. Scanning electron microscope and infrared spectrum result showed that different modification method exerted great influence on the structure of dietary fiber, but the degree of crystallinity was still the same; improvement was shown in the water holding capacity and oil holding capacity of dietary fiber after modification, and better adsorption capacity was also embodied to cholesterol, NO2- and sodium cholate, but little variation was shown in expansibility and cation exchange capacity. In general, superfine grinding by wet processing has best effect.(4)The dietary fiber of Passion Fruit which was modified by superfine grinding by wet processing was taken as the raw material, and the influence of five drying methods, such as forced air drying, microwave drying, vacuum freeze-drying, spray drying, spray freezing and drying, on the appearance and quality of raw material has been conducted. The result showed that different drying method will exert large influence on the appearance of dietary fiber; the dietary fiber processed by vacuum freezing-drying and spray freezing and drying was characterized by excellent water-holding capacity, oil-holding capacity, moisture absorption, as well as adsorption capacity to cholesterol, but the tap density was smaller; the dietary fiber processed by spray drying was characterized excellent liquidity; there was no large difference in expansive force and cation exchange capacity.(5)Construct high-fact mouse model with high-fat diet, feed the mouse by intragastric administration with wet-modified dietary fiber and common wheat bran for six weeks, and measure the effect of reducing blood fat, relaxing bowel, and liver protection function in high, medium and low dose groups. The result showed that apart from the blank control group, the activity of TC, TG, MDA, ALT and AST of the mice serum of positive control group, high dose group and medium dose group declined to some extent; the activity of HDL-C, GSH-Px, SOD increased to some extent; but the LDL-C of high dose group and medium dose group declined, and was significantly different from the model group, indicating that dietary fiber has sound function in reducing blood fact and liver protection, and certain relation was existed in effect and dose. Compared with the model group, the mice defecation of each intragastric group was not significant, but the fecal moisture increased significantly, indicating that such modified fiber also has certain effect in relaxing bowel.
