Cis-epoxysuccinate hydrolase is a kind of epoxide hydrolase and can added water molecules to the epoxy substrate selectively and produces 1,2-diol. And immobilized ESH can improve enzyme stability. Therefore, it is common in enzyme catalysis. L- tartaric acid is a natural organic acid and can be used in food additives and pharmaceutical industry. The paper mainly study the catalytic properties and kinetic properties of immobilized ESH, and study the production of ESH from agarose immobilized and the enzymatic production of L- tartaric acid and the clean production of L- tartaric acid by ion exchange.1. Study on the catalytic properties and fundamental kinetics of immobilized ESHComparison of free and immobilized ESH in catalysis of substrate, temperature,pH, and speed can effect stability, immobilization of ESH on temperature and rotational speed was improved. through the experiment. On the pH, the free ESH is higher than that of the immobilized ESH. The optimum reaction temperature was37℃, and the optimum pH was 8, and the optimum reaction speed was150r/min. Through experiments, the Vmax of free Vmax ESH is 2.595 mM/min and Km is 35.71 mM. The Vmax of Immobilized ESH is 1.637 mM/min and Km is 34.27 mM.2. Immobilized ESH enzymatic conversion of L- tartaric acid production of potassium sodium tartrateESH which is fixed in the agarose medium can producte L- sodium potassium tartrate. The optimum flow rate is 5 mL/L. It is investigated the catalytic ability of immobilized ESH. Immobilized ESH were eluted and regenerated, and can still maintain a certain catalytic ability.3. Enzymatic conversion of L- tartaric acid and application of ion exchangeUsing 001×7 cation exchange resin on the adsorption of Na +, L- sodium tartrate turns into L- tartaric acid. Calculated by the experiments, its static adsorption is 45.91mg/g resin, and its dynamic adsorption capacity was 50.93 mg/g. Cation exchange resin on L- 0.5 mol/L sodium tartrate adsorption effect is best. The method of ionexchange can producte L- tartaric acid cleanly.
