Procalcitonin is the precursor of calcitonin, a kind of glycoprotein has no hormone activity.In recent years, many studies have shown that the PCT was closely correlated with severe systemic bacterial infections and prognosis.Because of high sensitivity and specificity,it now becomes an indicator of clinical testing.So the quantitative detection of PCT is of great significance in clinical diagnosis and determination of the treatment program.In this paper,using the purified antigen in the laboratory,respectively immune rabbits and mice to prepare PCT polyclonal antibodies and monoclonal antibodies,couple with quantum dot,estabished a methodology of the detection for procalcitonin based on fluorescenceimmunochromatography.At the same time,prepare the strip and carry on the preliminary application. The results show that the test strip for PCT provide a linear response range from 0.15 to 120 ug/L and a limit of detection of 0.0075 ug/L,the recovery rate between 91% and 113%,the coefficient of variation(CV) of within-and between-run variation in the immunoassay system was<10%.In this paper,in comparison with PCT Elisa Kit from aboard on the market,the proposed strip for PCT has a lower limit of detection and the trend of test results are consistent. This paper established the high sensitivity, fast, convenient PCT test method has important significance for clinical detection of bacterial infection, and guide the rational use of antibiotics,for the development of PCT fluorescence diagnostic strip and related products provides an experimental basis.
