Sophorolipid(SLs)is an extracellular glycolipids of secondary metabolite by the non-pathogenic yeast.Firstly,this paper was enlarged the fermentation process of sophorolipid,in order to improve the total production of sophorolipid;then studied and contrasted the pharmaceutical activity on Hela cells of the sophorolipid and sophorolipid equipped with nano-hydroxyapatite;Finally studied the pharmacology active role on pathogenic bacteria of sophorolipid.Provided reference basis for large-scale application in the future.The main research content and results were as follows:1.Enlarged the fermentation process of sophorolipid.On the basis of optimized fermentation medium to enlarge the fermentation system of Wickerhamiella domercqiae Y2 A CGMCC 3798,studied its effect on sophorolipid total production.we use the method of fermentation system to 5 L,50 L fermentor to enlarge the fermentation process,the results showed that the process from the flask to 5 L fermentation tank in raising the output of sophorolipid total yield increased from the original 42 g/L to 84.5 g/L.Fermentation time was also greatly decreased,reduced from the original 196 h to 96 h,the reaction time shortened by half.Then the fermentation system was further expanded to 50 L,the sophorolipid yield was no significant effect,and both of the required fermentation time was flat,the total yield of sophorolipid from 83.8 g/L increased to 84.5 g/L.Analysized the reason of amplified by the fermentor process to improve the yield of sophorolipid,the content of oxygen in the fermentation broth was changed,and stimulated the growth and metabolism of yeast.2.The research of pharmaceutical activity conparerd SLs with nHA-SLs on cervical cancer cells.The effects of sophorolipid on cancer cells were investigated in vitro,the data showed the lactone diacetyl sophorolipid with good inhibitory effect on the proliferation of Hela cells,the survival rate of Hela cells was greatly reduced.Obvious changes have taken place in cell morphology,such as cell membrane bulging,volume decreased,cell swelling,cracking into pieces and collapse.By nano-hydroxyapatite loaded,compared with free sophorolipid,sophorolipid drug delivery time has been extended from 30 min to 14 h,nHA-SLs can continue to supply sophorolipid molecule to the tumor cells,which provided long uninterrupted supply for tumor cell therapy,and avoided the waste of sophorolipid drugs.This paper was researched the inhibition activity of sophorolipid for Hela cells in vitro,laid the foundation of sophorolipid as a new generation of anticancer drug.3.Pharmacological activity on pathogenic bacteria of sophorolipid.The hyphae was their vegetative in most fungal pathogens,this paper added different concentrations of sophorolipid and compared with Jinggangmycin,both of them had significant inhibition of Phytophthora infestans hyphae.With the increase of drug concentration in the medium,spore germination counted less and less,when the concentration was 10 mg/m L of sophorolipid in the PDA medium,the inhibition rate of spores which could reach 98.96%,when the concentration of Jinggangmycin was 100 ?g/mL,68.40% of spores could not survive.When the concentration was 10 mg/m L of sophorolipid in the PDA medium,the mycelial growth inhibition rate was reached 91.29%,and when the concentration was 100 ?g/mL of jinggangmycin,the inhibition rate was increased to 89.92%.This study showed that sophorolipid had good inhibitory effect on crop pathogens,and had excellent characteristics,such as no stimulation to respiratory tract,do not have harm to the skin and no environmental pollution and so on.
