Abstract/Summary:
Rice bran oil deodorizer distillate (RBODD) is the waste during the process of refining of rice bran, which includes fatty acid, sterol, VE, squalene and etc.As is well known, the sterol, VE and squalene have very high nutritional value that they can be widely used in food additives, cosmetics, medicines and so on. Therefore RBODD is of high value, how to make it to realize the effective utilization of resources, is of great significance.First of all, using GC-MS, we have analysed the main components of RBODD and identified content. Because the Fatty acids have content of about 75% in the RBODD. They affect extraction of other active substances. If we want to extract this active materials, we need to deal with free fatty acids, which helps us to separate squalene, VE and sterol in the system.The content of VE is very low, only 0.4%. It can be used as additional product in separation extraction process. At the same time we also analyzed the composition of fatty acid in RBODD. Oleic acid and linoleic acid are both accounting for 74% of the fatty acids total mass. The second is the palmitic acid which is 15% of the fatty acids total mass.Laboratory extraction process is divided into four parts:The reaction of methyl, complexation reaction, methyl distillation and column chromatography.After optimization, the best conditions were determined:20 g RBODD,40℃,170 rpm, adding methanol three times which each time adding 0.8 mL,add 2% dosage of diatomite lipase, reacting about 24 h, the conversion of FFA can be as high as above 95%. Then Sterol has good strong complexation ability to calcium chloride. The optimal reaction conditions were determined:70℃,300 rpm, complexation reaction about 2h.Through two complexing, sterol yield was 80%, the purity was 98.5%.We get the fatty acid methyl ester through methyl distillation.Eventually the yield of FAME was 90% and the purity of FAME was 98.5%.By column chromatography,we can get the yield of squalene and VE which was 87.8% and 80%,And the purity of squalene and VE was 94% and 92.5%.In this paper, the experimental process take amplification experiments The conversion rate of FAME can reach more than 95%. We can get the yield of squalene was 86%and the purity of squalene was 90% with Molecular distillation. Finally by column chromatography separation sterol and VE, eventually get the yield of sterol and VE were 83% and 80%, the purity of squalene and VE were 96% and 94%.
