| Capsaicin is the primary capsaicinoids the spicy material in the fruit of genus capsicum.The chemical structures of capsaicin is trans-8-methyl-N-vanillyl-6-nonenamide.Capsaicin with many special bioactivities is widely used in fields of agriculture,military,medicine,coating,etc.The high purity of capsaicin is needed in medicine specially.capsaicin with high purity was extracted from capsicum oleoresin and prepared with the combination method of aqueous two-phase extraction and two step column chromatography separation,capsicum oleoresin used as raw materials.In this paper,the research contents and results are as follows:(1)Capsaicin was extracted from capsicum oleoresin with ATPS.The property of forming aqueous two-phase and extracting results of capsaicin of aqueous two-phase systems formed by copolymers of ethylene oxide and propyl-eneoxide(EOPO(L31,L44,L35,UCON-HB 5100)and four different salts(Na2CO3,Na2HPO4,(NH4)2SO4,Na-3C6H5O7·2H2O)was studied.The UCON50-HB-5100/ K2HPO4 system of high extraction efficiency to capsaicin was choosen to study the effects of UCON50-HBO-5100 content,K2HPO4 content,p H of buffer,ethanol content,sample loading and extraction time on extraction efficiency of capsaicin.Morover,Box-behnken was used to determine an optimized aqueous two-phase system for the extraction of capsaicin from capsaicin oleoresin.The result was that in a 16.3%(w/w)UCON-HB 5100/10%%(w/w)K2HPO4,1% ethanol,at the pH of 4.35,sample loading of 0.2 g,extraction time of 60 min,95.5% of the capsaicin was recovered in the top phase.(2)The crude extract from aqueous two-phase extraction was performed preliminary purification by the nonpolar macroporous adsorption resin ADS-17.The optimum technological condition are as follows: 150 mL sample with the capsaicin concentration 84.93μg/m L,and pH 3 was loaded on the macroporous resin ADS-17 at the rate of 1.5 BV/h.Capsaicin was gradiently eluted with deionized water-30% ethanol aqueous solution 5BV-70% ethanol aqueous solution 5BV at the rate of 1 BV/h.As the result,the recovery and purity of capsaicin was 85%% and 51.5%,respectively.(3)The crude extract from aqueous two-phase extraction was performed preliminary purification by the nonpolar macroporous adsorption resin ADS-17.The optimum technological condition are as follows: 50 m L sample with the capsaicin concentration 85μg/m L,and pH 2 was loaded on the macroporous resin AB-8 at the rate of 2BV/h.Capsaicin was gradiently eluted with deionized water-30% ethanol aqueous solution 7BV-50% ethanol aqueous solution 6BV at the rate of 1.0BV/h.As the result,the recovery and purity of capsaicin was 89.2%% and 85.4%,respectively.The experiment used the combination method of UCON50-HB-5100/K2HPO4 /ethanol and two step column chromatography macroporous adsorption resin ADS-17 and AB-8 to obtain capsaicin with high purity. |
