China is rich in rice resource which is not only as the staple food,the people pay more attention on the healthy function of black rice with the improvement of people’s living standards.Traditional Chinese medicine says black rice tastes sweet and warm;It can lead the heart blood,make the spleen and stomach healthy,remove blood circulation.Researches have shown that black rice have many physiological functions,such as promoting higher plasma HDL cholesterol,improving iron deficiency anemia,resistancing stress response and promoting immune regulation.In this study,black rice was used as raw material through the optimizting of the extraction process of black rice pigments by single factor experiments and response surface methodology.We worked on the stability of extraction after the purification of flavonoids crude extract,which can used for the scientific development and utilization of black rice resources.The results showed that:(1)On the basis of single factor test,orthogonal test to optimize the extraction of black rice pigment technology,optimal conditions for the black grain of 80 mesh,the extraction agent of 0.5%citric acid and 70%ethanol solution,extraction temperature is 70 ℃solid-liquid ratio of 1:3,50%ethanol concentration,extraction time 30min,to give black rice pigment color price of 7.51,by optimization of verification test results extraction rate of 13.77%.(2)Static adsorption and desorption experiments shows that AB-8 resin was selected to purificating pigment.When the value of the sample concentration is 1.5mg/mL,pH is 2.0,the sample flow rate is 1mL/min,AB-8 resin adsorption of pigment effect is best,saturated adsorption amount of the resin is 0.0147g/mL.Dosage of eluent water is 75ml,Eluent pH1.5,the eluent is 130ml 75%ethanol,elution flow rate is 1.0mL/min we got the highest desorption rate.Color value promotedby 15%after purification.
