Rice protein is one of the major sources of plant protein people intake. In plant protein, rice protein is regarded as the optimal plant protein. Amino acid composition of rice protein ratio is reasonable, close to the world health organization decided the best ratio of protein amino acid pattern. Main rice protein are glutelin, the glutelin are large molecular weight, large amounts of thiol and disulfide were existed in glutelin. These features make rice protein poorly soluble in water, the serious influence rice protein application in food industry.Therefore, how to improve the efficiency of the extraction of rice protein is an important scientific problems.This work was composed of three parts including exploring the extraction method of rice protein, enlarging the extraction at the laboratory-scale, and the pilot-scale design for extraction of rice protein. The main work, results and conclusions are presented as follows.1. Firstly, comparative literature and common several kinds of plant protein extraction, and designed several extraction method are doing experiments,determine the total rice protein, salt-soluble protein and salt-insoluble protein extraction method finally. The result showed that based on the strategy of 50 m M Tris-HCl(p H7.2)?2%(w/v)SDS?0.6%(v/v)?-Mercaptoethanol?4M Urea is a relatively efficient totalriceprotein extraction method, extraction of protein concentration is 3,050±94.77mg/m L; based on the strategy of 1mol/L Na Cl?30m MTris-HCl(p H8.0)is a relatively efficient salt-soluble protein extraction method, extraction of protein concentration is 750.00±50.92mg/ml; based on the strategy of 55 m M Tris-HCl(p H6.8)?2%(w/v)SDS?10%(v/v)glycero?11mg/ml DTT rice flour extraction directlyis a relatively efficient rice protein extraction method, extraction of protein concentration is 1,126.66±73.40mg/ml. This extraction approach is reliable and easily for handing, equipment requirements not complicatedand it is suitable to be enlarged for preparing rice protein.2. The preparation of rice protein was enlarged 100 times. The concentration ratio of two experiments is 99.26%,99.09%,99.08%, this shown that the method has good repeatability. Therefore, product yield are well kept, for the pilot design provides a reliable experimental parameters.3. A pilot-scale design for the extraction of rice protein was carried out based on the laboratory-scale extraction parameters. Annual output of 100 tons of products factory design was completed.
