Calcium amino acid chelate has the advantages of high absorption rate, small side effects and so on, and the main component of amino acid chelated calcium calcium products is calcium carbonate in the city sale, calcium preparations whose main component is glutamate chelate calcium which made from egg shell with biological activity are rarely reported. In this paper, the effect of ultra fine powder and coarse powder on the chelate rate of glutamic acid chelate was studied, the calcium glutamate chelate tablets and soft capsule ingredients and process were optimized, the stability of soft capsules and tablets were evaluated, The main results are as follows:(1)All the qualities of eggshell powder were the best when pulverizing time was 4 min; chelating rate of glutamate chelating calcium increased by 12.57%, which prepared with eggshell ultra micro powder compared with coarse eggshell powder, at the same time, the utilization rate of glutamate was improved. The optimal conditions optimized by the response surface for the preparation of glutamate chelating calcium were as follows: the added mass of glutamate was 21.40 g, the pH value was 7, the time of chelating was 59 min, and the temperature of chelating was 60℃. Under the optimal conditions, chelating rate of glutamate chelating calcium was 81.12%, which prepared by eggshell ultra micro powder.(2)Through the screening test and response surface optimization test, the results were as follows: the granulated degree of glutamate chelating calcium powder is 120 mesh, the optimal content of ingredients were as follows: glutamate chelating calcium was 41%, amount of filler microcrystalline cellulose + corn starch was 41%, addition of disintegrating agent hydroxypropyl methylcellulose was 20%, addition of wetting agent micro-powder silica gel was 1.95%.(3)Through the screening test and response surface optimization test, the results showed that degree of glutamate chelate calcium powder granulated was 120 mesh, optimal content formula of soft capsule: the ratio of glutamate chelating calcium powder to the diluent was 1:4, adding amount of suspending agent PEG4000 was 10%, mass ratio of diluent soybean oil to PEG400 was 1:1, adding amount of wetting agent Twain-80 was 2%.(4)Normal quality inspection and stability test for preparation of glutamate chelating calcium agent and soft capsule were made, the results showed that, the indicators of tablets and capsules were in line with the provisions of the <Chinese Pharmacopoeia> in 2010 edition, the average content of glutamate chelating calcium were 196 mg /tablet and 105 mg / particle respectively. In the accelerated conditions, quality of the tablet and capsule remained stable and good stability.
