Abstract/Summary:
Amino acids are the umbrella name of the organics which have amino and carboxyl.They are the foundation of protein.An amino acid is a kind of organic compounds with containing at least of one amino which is acid and one carboxyl which is alkaline.As a kind of necessary nutrients,amino acids not only are used synthesis protein,but also the material of lots of animal feed and food.In clinical medicine,amino acids are used for the prevention and treatment of various and in some chemical products is also a kind of important active ingredients.Although amino acids play an important role in the human body,that is not to say the more absorb about it the better in our daily lift,too more amino acid can increase the burden of liver and kidney;the longtime excess may cause diabetes.In recent years the methods of amino acids determines have Kjeldahl method,formaldehyde titration,spectrophotometry,thin layer chromatography,paper chromatography,liquid chromatography,meteorological chromatography,meteorological chromatography,electrochemical analysis,automatic amino acid analyzer.All of the methods described above have the disadvantage such as higher cost and low sensitive.Compared with traditional measurement method,fluorescence spectrometry has the advantage such as short test time,simple operation,low cost,highly sensitive and so on.It is a meaningful thing to establish a method for amino acids determination.This paper was mainly divided into three main substances:1.In this article,we classify different amino acid and summarize the useful in our daily life,compare with the disadvantage and advantage between the traditional methods and fluorescence spectrometry,and introduce the basic principle of fluorescence spectrometry determination of amino acids.2.Establish three fluorescence spectrometry methods for amino acid determination:(1)A novel method for determination of glycine was established.In a B-R buffer solution(pH = 9.6),the fluorescence intensity of fluorexon,which is quenched by Ni2+ can be enhanced after adding a series of glycine,and the increase has a good linear with the glycine.Based on this discovery,we established a new fluorescence spectrometry method for the detection of glycine.(2)In the Clark-Lubs buffer(pH=2.8)solution with surfactant CTMAB,the fluorescence intensity of Salicyl fluone is significantly quenched by Cu2+ due to the complexation reaction.While adding a certain amount of methionine into the solution,the fluorescence of the system gradually recovered.Based on this,a fluorescence spectrometry method for detection of methionine was established.(3)This study presents a simple,accurate method for the determination of proline.In a Kolthoff buffer solution(pH=10.6)with the cationic surfactant coexistence,the fluorescence intensity of calcein is quenched by Cu2+ but can enhanced after adding a certain amount of proline.The fluorescence intensity increment shows a good liner with the concentration of the proline.Based on the phenomenon we establish a method for determination of proline.3.Through investigating the fluorescence intensity variation between the primitive system and after adding quencher,we calculate the reaction mechanism and ration.We calculated the ration and stable constants of compounds by spectrophotometry method.
