Uronic acid-containing polysaccharides(UACPs)are important nutriceuticals in aquatic products.In this study,a UACP analysis method has been established using HPLC-MS/MS,and was applied to the determination of UACPs compositions in 13 aquatic products.The products of hyaluronic acid(HA),chondroitin sulfate(CS),heparin(HP)and AGSP with backbone of ?4)-Glc A(1?2)-Man(1? repeating units after acid hydrolysis and 1-phenyl-3-methyl-5-pyrazolone(PMP)derivatization were analyzed.10 characteristic ion pairs were chosen for the MS detection,and the collision energy(CE)and declustering potential(DP)for every characteristic ion pair was optimized,and then acid hydrolysis-LC/MS/MS method for determination of the UACPs was established.In this study,polysaccharides were isolated by digestion with trypsin and papain,and analyzed by the hydrolysis-LC/MS/MS method.As shown by the results,HA,CS,HP and an unknown UACP containing repeated disaccharide of hexuronic acid linked to hexose were observed in the viscera of Ctenopharyngodon idellus.CS,HP and minor HA existed in the viscera of Cyprinus carpio.CS was detected as the major UACP in viscera of Larimichthys crocea and Trichiurus lepturus.The analytical results of different tissue parts of Saxidomus purpurata,Cyclina sinensis,Chlamys farreri,Neverita didyma and Natica janthostomoides revealed that there were differences of the distribution of UACPs in different organs of shellfishes.There were some similarities of the distribution of UACPs between two kinds of shellfishes with close genetic relationship,such as Saxidomus purpurata and Cyclina sinensis,and Neverita didyma and Natica janthostomoides.The comparison of UACPs in adductor muscle and viscera between female and male Atrina pectinata showed no significant differences between female and male samples,but indicated significant differences in UACP composition between viscera and adductor muscle.This study also suggested that the HPLC–MS/MS method provided a power tool to compare proportions of UACPs in question samples.What's more,the analytical results of scallop adductor,shredded squid tube,and cod fillet showed that CS existed in all of the 3 ready-to-eat seafood products,and a little HA was observed in the cod fillet.This study revealed the UACPs in the 4 fish viscera,6 edible shellfishes and 3 ready-to-eat seafood,which provided the scientific basis for the development and use of these aquatic products and promoted the understanding of nutrients in the animal-derived aquatic products.
