Fluorescence analysis (FS) has been one of the subject of numerous investigations in modern chemical analysis because of its attractive advantages such as fast analysis, high separation efficiency, little consume and lower limit of detection and so on. Aromatic amino acid such as tryptophan (Trp)、Tyrosine(Tyr) and Phenylalanine(Phe) has been used in fluorescent detection and drug analysis because of its fluorescent properties. In this paper, application of three kinds of aromatic amino acids(Trp,Tyr and Phe) by means of fluorescence analysis, the concrete works are as follows:Firstly, the effects of three different kinds of system on the preparation of L-phenylalanine methyl ester hydrochlorides were studied using L-phenylalanine as material. Besides, three kinds of amino acid methyl ester hydrochloride were obtained under the optimum reaction conditions. Furthermore,7-(menthoxyacetic acid) coumarin was synthesized by a two-step reaction from 7-Hydroxycoumarin, which further reacted with L-phenylalanine methyl ester hydrochloride to obtain a new type of coumarin derivatives N-acetylcoumarin (oxygen-7-coumarin)-3-phenyl-2-amino-methyl propionate by acylation reaction. The structures were characterized by FT-IR and 1HNMR. Finally showed that N-acetylcoumarin (oxygen-7-coumarin)-3-phenyl-2-amino-methyl have good recognition ability about Pb2+. It has potential application value in ion detection and recognition.Under the condition of different temperatures, the interaction of hydrocortisone (Hyd) and Nystatin (Nys) with tryptophan (Trp), tyrosine (Tyr) and phenylalanine (Phe) were studied in Tris-HCl buffer solution at pH 7.4 via fluorescence spectroscopy.The type of interaction, points of binding,binding constant and the type of interaction foce been studied.It turned out that when the small drug molecules(hydrocortisone) Was added to the Amino acids (tryptophan,tyrosine and phenylalanine),the fluorescence is quenching. The quenching occurred through the formation of the complexes of Hyd with each aromatic amino acid in a molar ratio of 1:1, Hydrogen bond and vander Waals force may play a major role in the reaction of Hyd with Trp and Tyr, whereas hydrophobic interaction should be responsible for the binding of Hyd with Phe, Hydrogen bond and vander Waals force may play a major role in the reaction of Nys with Trp and Tyr, whereas hydrophobic interaction should be responsible for the binding of Nys with Phe.
