Herbicide plays an important role in agricultural production. With the increase of weeds resistance and the environment protection requirement, it has an urgent need to develop more effective, low toxicity and environmental friendly herbicides. Because the limitation of present known herbicides mechanism and targets, it has become the bottleneck of herbicide development, so searching for new targets and determination of the target protein’s structures are the primary stage of target structure-base herbicide design and development.According to the demand of new herbicides, we propose a structural genomic study on novel potential prottein kinase-based herbicide targets. Refer to the successful experience of medicine development, new kinase genes from Arabidopsis thaliana and Qryza sativa genome were functional analyzed by comparative genomics method. By the strategy of sequence alignment and the comparison of secondary structure, we selected 100 protein kinases as the potential new herbicide targets. Using molecular cloning technology, we amplified 29 genes by the method of polymerase chain reaction (PCR), and constructed 24 recombinant plasmids.
