| Studies have shown that polysaccharide is the main active component of Auricular auricular,it has a wide range of pharmacological effects such as antiaging,fall blood sugar,antibacterial etc,the development of Auricularia auricula polysaccharides(AAP)is promising in the field of application.This paper studi es the black fungus liquid fermentation process optimization and optimization of the extraction process of polysaccharide and its antibacterial and antioxidant activity.The main findings are as follows:(1)On the basis of single factor test and orthogonal design optimization,this paper researched the effect of carbon,nitrogen,trace elements proportion,liquid volume and other conditions on liquid fermentation,From the study,the optimum conditions for liquid fermentation: 3% glucose,0.5% yeast extract,0.1% potassium dihydrogen phosphate,0.05% magnesium sulfate,250 mL shake bottled liquid volume 90 mL,speed 140 r / m.(2)On the basis of single factor experiment,the orthogonal experiment an d the response surface optimization design,to study the effects of water extrac tion and alcohol precipitation extraction temperature extraction of black fungus polysaccharides,solid-liquid ratio,extraction time,extraction times,the establishment of a total of polysaccharide Multiple factors affect quadratic regression equation,and so determine the AAP’s optimum extraction conditions were: extraction temperature 61 ℃,extraction time 2h,1:23 for the ratio of solid to liquid,extracted 3 times.(3)The result of AAP’s antimicrobial experiment: The AAP has antibacter ial effect on E.coli and Staphylococcus tetragenus,on the basis of scanning electron microscope and live-dead staining test results,we can learn that the in creasing concentration of AAP,the better inhibitory effect.(4)This experiment to test the antioxidant activity of BHT,AAP and VC.the total restoring force: BHT﹥VC﹥AA;DPPH Free radical scavenging activity : VC﹥AAP﹥BHT;The hydroxyl radical scavenging ability: VC﹥AAP﹥BHT. |
