| Endocrine disrupting compounds(EDCs)is a kind of chemicals who affect the endocrine system, reproduction and development, the nervous system, immune system and have carcinogenic. Most of them are synthetic and human production and emissions into the environment with the ptoduction and life of human. Even low concentrations in the environment, their can cause endocrine disruption of biological effect,so their has become the focus of international scholars. In recent years, as the result of the extensive use in the daily life and the cultivation, the emissions of different kinds of estrogen is increasing, this situation has caused worldwide attention.As the concentration of EDCs is very low in water environment, it is difficult to measure accurately. Normally, Solid Phase Extraction(SPE), Solid Phase microextraction(SPME), Dispersive liquid-liquid microextraction(DLLME) are appalied in pretreatment to enrichment of EDCs, after that high performance liquid chromatography(HPLC), gas chromatography(GC), gas chromatography mass spectrometry(GC-MS), high performance liquid chromatography tandem mass spectrometry(HPLC/MS/MS) are used to analysis. Although these method can used to measure trace amounts of EDCs in water environment accurately, but they are timeconsuming, high equipments, expensive and can’t meet the requirements of rapid detection and mass samples daily. Enzyme-linked immunsor bent assay(ELISA), who is based on antibody and antigen specific reaction, is one of the important method used to detect the levels of EDCs in biological samples, as its detection speed, high sensitivity and low price. Based on this, this study using ELISA detect Estradiol(E2) which is the most typical environmental endocrine disruptors, and then use this method to detect sewage in a sewage treatment plant in Xi’an. The results showed that:the recovery of the wastewater sample is 73.33% to 115.5%; The intraday precision for100ng/L, 200ng/L, 300ng/L is 8.36% to 9.05%, and its interday precision is 8.77% to 9.63%, the results conform to the requirements of trace analysis; under the condition of not for enrichment of water sample the limit of detection of ELISA is 29ng/L. Compare with traditional HPLC method, ELISA method have the advantage of limit of detection lower, rapid and cheap; But it also easy influence by similar substance.We using solid-phase extraction technology to extract and high performance liquid chromatography(HPLC) to detect five kinds of typical EDCs in urban sewage samples. To understanding the distribution and removal characteristics and the influence of seasonal weather on the distribution and existence of five kinds of EDCs in different structures, we choose 6 sampling points between different structures in a sewage treatment plant in Xi’an to carry on the long-term monitoring. What we want is to build the base of study which is the trandition and transform of EDCs in sewage. Through the long-term measurement results of 5 types of EDCs in six sampling point of Xi’an a sewage treatment plant, we study that: the concentration of 5 kinds of EDCs in the inlet and out of the water were respectiveiy in the range 119.88~38.55μg/L and 54.21~5.42μg/L, estriol(E3) as the most in the inlet and out of the water of five kinds of EDCs, in and out of the water acounted for 76.38% and 71.06%of the total respectively; the best removal effect of five EDCs by sewage treatment plant is estradiol(E2) and ethinyl estradiol(EE2). The results of testing five kinds of EDCs in water of different structures showed that: the degradation of five kinds of EDCs removal effect mainly in the stage of biochemical treatment.Different sampling time show that the content of EDCs in the inflow has the certain regularity, and part of the degradation of EDCs removal has certain correlation with the temperature and season. |
