This is the Jilin modern agriculture key research projects-mussel products processing key technologies and integrated product development(20150204034NY) and part of the research project of Changchun modern agricultural technology development support program-mussel products processing key technology research and comprehensive utilization of resources(13NK13)Mussel is a generally aquatic animal of which functional factoris mainly mussel polysaccharide. It was shown that the polysaccharide had healthcare functions such as blood cooling, anti-coagulation, lipid-lowering and anti-aging. What’s more, the polysaccharide could also increase phagocytosis ability of macrophage and inhibit development of cancer cells. In Songhua River basin in Jilin province, mussel resource is rich with yield of 3000-4000 t annually and cheap fishing price of RMB 1-1.2/kg. However, most of mussels are discarded as garbage except little of them as fodder, resulting in environment pollution and resource waste. As a result, it is significant for enhancing economic benefit and decreasing environment contamination if high additional mussel products were developed after artificial pearl culture. This paper uses wild mussel byproduct-mussels tears and visceral tissue as raw material,use enzymatic hydrolysis assisted by high-voltage pulsed electric field to extract polysaccharide、Sevage method remove free protein、DEAE-cellulose column and Glucan G-100 gel column chromatography to separate and purify the coarse polysaccharide, and the study on the antioxidant activity of polysaccharide,and provides a theoretical basis for the comprehensive development and utilization of efficient extraction and mussel byproduct of polysaccharides. The main contents and conclusions are as follows:1.Using papain hydrolysis and enzymatic hydrolysis assisted by high-voltage pulsed electric field to extract polysaccharide efectively,studied influence of pH, water-feed ratio, reaction time, reaction temperature, electric field strength, the number of pulses and other factors on mussel polysaccharide yield. The results show that, hydrolysis assisted by high-voltage pulsed electric field to extract polysaccharide has the advantages of high yield, short time-consuming.2. Enzymatic hydrolysis assisted by high-voltage pulsed electric field to extract polysaccharide,the the quadratic regression equation on the relationship of yield (Y) and the electric field intensity (Xi), the number of pulses (X2) and the hydrolysis time: Y=-30.2578+1.67015X,+1.062125X2+6.667X3-0.00775X1X2-0.038X,X3-0.12X2X3-0.02479X,2-0.03369X,2-1.309X32Through the analysis of variance, the optimal parameters of polysaccharide extraction are field strength 30.96kv/cm, the number of pulses 9.22, reaction time 1.67h, corresponding mussel polysaccharide yield 6.08%;various factors on the impact of mussel yield of polysaccharides order of significance:the electric field strength> hydrolysis time> pulses.3. Free proteins in mussel crude polysaccharside were removed by Sevage method,and get the protein polysaccharides;Use DEAE-cellulose column and Glucan G-100 gel column chromatography to separate and purify the coarse polysaccharide; Use MWCO were 3500 Da,8000-14000 Da dialysis bag dialysis to remove low molecular impurities to afford the pure polysaccharide AWPS-1 and AWPS-2, its yield was 5.42% and 1.96%, respectively.4. Study of mussel polysaccharide sensory characteristics and solubility studies show:mussel polysaccharside was colorless laminar crystals,it will became light yellow in air completely for a long time,with slight earth smell,the polysaccharside could dissolve in hot water easily, however it could hard dissolve in cool water, the polysaccharside couldn’t dissolve in many organic solvent.5. Polysaccharides, total sugar contents of pure polysaccharide AWPS-1 and AWPS-2 were73.54%,97.79%,95.33%; protein content of crude polysaccharide was 2.24%; pure polysaccharide AWPS-1 and AWPS-2 does not containing sugar residues, free amino acids and proteins.6. Mussel polysaccharides, pure polysaccharide AWPS-1 and AWPS-2 have the superoxide anion radical scavenging, hydrogen peroxide scavenging, hydroxyl radical scavenging capacity, reducing power and DPPH-scavenging and with increasing concentration of polysaccharides,antioxidant capacity enhance and antioxidant capacity was positively correlated with the concentration of the polysaccharide.Innovations:For the first time using enzymatic hydrolysis assisted by high-voltage pulsed electric field to extract mussel byproduct-mussels tears and visceral tissue as raw material, its high yield, short time-consuming, turning waste into treasure, higher value added products, for efficient extraction of polysaccharides provide a mussel seed new approach for resource utilization mussel open a new way.
